このプロトコルは、スフェロイドの三次元腫瘍を構築する標準化された方法を実証しました。そして、この方法論は、3D腫瘍構築物の高回路および高コンテンツ分析を提供しました。腫瘍性スフェロイド構築の標準的な方法とハイスループットイメージングおよび分析システムを使用することにより、3次元スフェロイド上に形成される薬物検査の有効性と精度を劇的に向上させることができます。このプロトコルでは、例としてNCI-H23スフェロイドを治療するためにAMG510を使用しました。実験から、腫瘍スフェロイドに対する癌標的薬の有意な効果を観察することができました。はじめに、100マイクロリットルの付着防止試薬をU字型のウェル底部を有する48ウェルプレートの各ウェルにピペットで入れ、10分間保持する。10分後、コーティング試薬を吸引し、滅菌PBSで2回洗浄します。培養プレートを37°Cのインキュベーターに入れ、5%二酸化炭素を含む加湿空気中で使用時まで放置する。顕微鏡下で細胞を観察します。次に、T25フラスコで培養した細胞をPBSで2回洗浄して培養液を除去し、増殖した細胞を1ミリリットルの0.25%トリプシンEDTAで摂氏37度、5%二酸化炭素のインキュベーターで1〜2分間処理します。顕微鏡で細胞の形状を確認します。次いで、T25フラスコ内で使用済みのトリプシンEDTA懸濁液を吸引し、4ミリリットルの新鮮な培地で細胞を洗浄することにより、トリプシン処理を停止した。すべての懸濁液を15ミリリットルのチューブに移し、1ミリリットルの新鮮な培地を使用して残留細胞を洗い流し、チューブに追加します。細胞を186.48 Gで室温で5分間遠心分離し、上清を廃棄します。10ミリリットルの新鮮な培地を細胞ペレットに加え、細胞が均質な懸濁液になるまで静かにピペットで移します。0.1ミリリットルの細胞懸濁液を新しい遠沈管に吸引します。0.9ミリリットルの新鮮な培地を加えてから、懸濁液をよくピペットで移します。細胞計数のために10マイクロリットルの細胞懸濁液を抽出する。このプロセスを1〜2回繰り返し、平均値を取ります。懸濁液を希釈して、ミリリットルあたり50, 000セルの最終播種密度に達する。次に、200マイクロリットルの細胞懸濁液を48ウェルU底プレートの各ウェルに加える。シーリングフィルムをプレートに巻き付け、119.35 Gで室温で5分間遠心分離します。遠心分離後、保護フィルムを引き剥がし、井戸を囲む水路に5〜8ミリリットルの滅菌水を加えます。プレートを摂氏37度で5日間インキュベートします。この期間中は水路に水を交換または補給せず、次の5日間は細胞の凝集を観察してください。ゲルの埋め込みは、マイナス20°Cの冷蔵庫から冷凍ジェルを取り出した後、実験中ずっとアイスボックスに置きます。細胞スフェロイドを顕微鏡で観察します。ゲルの包埋を開始する前に、スフェロイドの状態を再度確認する必要があります。150マイクロリットルの培地を慎重に取り出し、予冷したピペットチップをウェル内およびウェル内を移動させながら、ウェルの壁側から液体ゲルをゆっくりと追加することにより、各スフェロイドをゲルに埋め込みます。5分間待ってから、ゲルが均等に広がらない場合は、10マイクロリットルのピペットチップでゲルを静かにピペットでピペットします。各ウェルには、腫瘍スフェロイド、25マイクロリットルの3.5ミリグラム/ミリリットルのゲル、および50マイクロリットルの完全培養培地が含まれています。コントロールにも75マイクロリットルの培地を追加します。ヒドロゲル化が完全に完了するまで、プレートを摂氏37度で30分間インキュベートします。ゲル化状態を顕微鏡で確認する。各サンプルに125マイクロリットルの新鮮な培地を重ね、スフェロイドをさらに7〜10日間培養します。それぞれ4〜6個のウェルを持つスフェロイドのグループを準備し、分析用に少なくとも3つを選択します。製造元の指示に従って薬を溶解し、DMSOで100倍の作業溶液を調製します。ポジティブコントロールとして0.1%DMSOを使用し、各ウェルに125マイクロリットルの薬物処理培地を追加します。プレートを5%二酸化炭素を含む加湿空気中で摂氏37度のインキュベーターに戻します。この段階で、各ウェルは腫瘍スフェロイド、25マイクロリットルの3.5ミリグラム/ミリリットルのゲル、および175マイクロリットルの培地を含む。コントロールには200マイクロリットルの培地が含まれています。メーカーのガイドラインに従って、alamarBlueアッセイキットを使用してスフェロイドの生存率を測定します。各ウェルから100マイクロリットルの上清培地を新しい試験プレートに吸引する。その後、マイクロプレート光度計を用いて生存率を測定する。スフェロイドをゲルに埋め込んだ後、1日目、4日目、7日目、および10日目に測定します。80マイクロリットルの新鮮な培地を培養プレートの各ウェルに加える。次に、さらに100マイクロリットルの薬物処理培地を交換します。井戸にアラマーブルーの残骸がないことを確認してください。イメージング前に100マイクロリットルの培地を吸引し、プレートをステージに置きます。スフェロイドのデジタル画像は、10倍の対物レンズを備えた自動顕微鏡を使用して取得します。顕微鏡は、これらのスフェロイドを自動的に焦点を合わせ、集中させることができます。自動イメージングを待ちます。各回転楕円体について4つの画像が取得されます。統合された画像が形成され、ハイコンテントイメージングシステムに接続されたソフトウェアで処理されます。画像パッチプロセスボタンをクリックして、ソフトウェアに統合された画像を選択します。U netモデルを選択し、コンバージョン率を入力します。画面下部のクリックで画像処理を開始します。直径、周囲長、粗さのデータを表計算ソフトウェアに保存します。最後に、100マイクロリットルの新鮮な培地に薬を加え、プレートを5%二酸化炭素を含む加湿空気中で摂氏37度のインキュベーターに戻します。異なる濃度のAMG510で処理され、高含有量顕微鏡によって自動的に撮影されたNCI-H23細胞スフェロイドの明視野画像をこの図に示します。列は異なる日を表し、行は異なる薬物濃度を表します。結果には、各条件の3つのスフェロイドが含まれます。AMG510処理サンプル群の腫瘍スフェロイド生存率を、1日目、4日目、7日目、および10日目に測定した。濃度勾配を有するサンプルの末端細胞生存率をこの図に示す。腫瘍スフェロイドの直径は、1日目、4日目、7日目、および10日目に測定されました。スフェロイド成長率は、元の体積に対する末端体積として定義され、スフェロイド直径を使用して計算されました。スフェロイド成長阻害比は、体積に対して定義し、スフェロイド直径を用いて計算した。腫瘍の粗さは、1日目、4日目、7日目、および10日目にソフトウェアによって測定され、腫瘍スフェロイドの浸潤性を示しました。回転楕円体の周囲は、ディープラーニングアルゴリズムを使用してソフトウェアによって特定および描画されました。次に、焦点面の回転楕円体領域を画像Jで測定し、これらのデータに基づいて過剰周長指数を計算しました。このメッセージは簡単に理解できますが、サンプルは慎重に処理する必要があります。
