1. SAA的建设

2. 麻醉剂暴露前

当前版本的SAA依赖于商业蒸发器，因此毒理学研究仅限于挥发性麻醉剂。如果与其他挥发性物质一起使用，则在校准输出后，可以在“标签外”使用蒸发器。或者，可以应用不同的挥发性物质汽化方法，这需要专门的测量来滴定药物浓度，如前所述25。
除了流量指示器外，没有警报（即，如果油箱是空的，则通过SAA的流量将被中断）。根据使用的强度，SAA可能需要清洁，拧紧并可能更换Tygon管。我们在 7 年的使用中两次对原始 SAA 进行了“维护”。
这种麻醉果蝇的方法允许在高通量系统中使用果蝇研究人员可用的遗传工具箱。不同种群（例如，基因型，年龄，性别）的多个苍蝇队列可以同时暴露于相同的麻醉剂浓度和适合手头研究问题的所需载气（空气，O 2，N2 O，惰性气体）组合。
我们在这里表明，SAA对于揭示 ND2360114 飞系中异氟醚毒性的恢复力的意外变化是有用的，并且标准实验室飞线对AP的响应性不同。
SAA可用于研究其他挥发性有机化合物（VOC）对昆虫（例如蜜蜂）的影响。对于蒸气压接近挥发性麻醉剂的挥发性有机化合物（异氟烷：20°C时为240mmHg），可以使用传统的汽化器，但必须校准输出。用于地氟醚的商用蒸发器是加热的，可能提供额外的灵活性。

附带麻醉效应（即不能立即观察到但可能具有延迟行为后果）的存在被普遍接受，但对其机制和危险因素的理解仍然很初步1，2。它们的延迟表现和微妙性限制了在合理的时间范围内以可接受的成本在哺乳动物模型中可以研究的潜在重要变量的数量。果蝇（黑腹果蝇）在神经退行性疾病3 和毒理学筛查4 的背景下具有独特的优势，迄今为止尚未应用于麻醉附带效应的研究。 
我们开发了连续麻醉阵列（SAA），以促进果蝇在麻醉药效学和药物遗传学研究中的使用。SAA的一个关键优势是同时暴露于多个队列的相同实验条件。当与果蝇的实验灵活性相结合时，SAA的高通量允许以哺乳动物模型中不可能的规模探索生物和环境变量。 
原则上，SAA 只是一系列连接的麻醉位置（由 50 mL 小瓶制成的腔室），载气通过这些位置输送挥发性药物。该系统的第一个腔室包含蒸馏水，载气通过蒸馏水加湿（苍蝇对脱水敏感），并通过一个简单的流量指示器终止，指示流经系统的气体流量。放置在连接管开口上的细网将腔室分开，以防止苍蝇在腔室之间迁移。“串联”位置的数量受到对非加压气流（管道、网）的阻力的限制。 
我们在之前的出版物5中描述了该SAA原型的动力学。尽管SAAs之间的确切药代动力学特性会有所不同，但经过实验测试的相关基础知识如下：（i）1.5-2 L / min的初始流量在2分钟内将所有腔室（总体积为±550mL）与所需的麻醉剂浓度平衡;（ii） 输送到腔室的麻醉蒸气浓度在第一个和最后一个位置之间没有明显变化，因为单个腔室气体体积（50 mL）中所含的麻醉剂量远远超过任何数量的苍蝇所吸收的量;（iii）一旦腔室平衡，可以减少载气流（50-100 mL / min或更低），以避免浪费和环境污染（挥发性麻醉剂具有温室气体特性）。保持蒸汽稳态浓度所需的最小流量主要取决于SAA的泄漏性，因为苍蝇对蒸汽的吸收可以忽略不计。在这些标准条件下（2%异氟醚和1.5 L / min载气流），在3-4分钟内在阵列的所有位置麻醉（即不动），位置之间差异不明显。VGA可以施用几分钟到几小时，我们的典型暴露范式在15分钟到2小时的范围内。为了冲洗系统，关闭蒸发器，并保持流量以交换大约 10 倍的阵列体积（1.5 L/min，持续 5 分钟）。麻醉剂消除的速度会随着设定的流速而变化。
