L. paraplantarum A FT-259 é uma bactéria Gram-positiva, em forma de bastonete, produtora de bacteriocinas com atividade antilisterial e com alto potencial probiótico20. Son et al.24 demonstraram previamente a capacidade imunoestimulante e antioxidante de cepas de L. paraplantarum. Além disso, possuem grande potencial probiótico, com propriedades como estabilidade em condições gástricas e biliares artificiais, suscetibilidade a antibióticos e liga?...

Para serem definidos como probióticos, os microrganismos adicionados aos alimentos (ou suplementos) devem ser consumidos vivos, ser capazes de sobreviver durante a passagem no trato gastrintestinal do hospedeiro e atingir o local de ação em quantidades adequadas para exercer efeitos benéficos 1,2,7.
O crescente interesse nos probióticos deve-se aos diversos benefícios à saúde humana que conferem, como a estimulação do sistema imunológico, a redução dos níveis séricos de colesterol e o aumento da função de barreira intestinal por atuar contra micróbios nocivos, bem como seus efeitos benéficos no tratamento da síndrome do intestino irritável, entre outros 2,3. Além disso, vários estudos têm demonstrado que os probióticos podem afetar positivamente outras partes do corpo humano, onde comunidades microbianas desequilibradas podem causar doenças infecciosas 3,4,5.
Para que os probióticos sejam terapeuticamente eficazes, o produto deve conter entre 10 6-107 UFC/g de bactérias no momento do consumo6. Por outro lado, o Ministério da Saúde italiano e o Canadá estabeleceram que o nível mínimo de probióticos nos alimentos deve ser de 10a 9 UFC/g de células viáveis por dia ou por porção, respectivamente7. Considerando que altas cargas de probióticos são necessárias para garantir que eles terão efeitos benéficos, é essencial garantir sua sobrevivência durante o processamento, armazenamento de prateleira e passagem pelo trato gastrointestinal (GI). Vários estudos têm demonstrado que a microencapsulação é um método eficaz para melhorar a viabilidade global dos probióticos 8,9,10,11.
Nesse contexto, vários métodos têm sido desenvolvidos para a microencapsulação de probióticos, como spray-drying, liofilização, spray-chilling, emulsão, extrusão, coacervação e, mais recentemente, leitos fluidizados11,12,13,14. A microencapsulação por spray-drying (SD) é amplamente utilizada na indústria alimentícia por ser um processo simples, rápido e reprodutível. É de fácil escalonamento, com alto rendimento de produção e baixa demanda energética11,12,13,14. No entanto, a exposição a altas temperaturas e baixo teor de umidade pode afetar a sobrevivência e a viabilidade das células probióticas15. Ambos os parâmetros podem ser melhorados para uma determinada linhagem, determinando-se os efeitos da idade e das condições de pré-adaptação da cultura e otimização das condições de spray-drying (temperaturas de entrada e saída, processo de atomização) e da composição encapsulante 8,14,16,17,18.
A composição da solução encapsulante também é um fator importante durante a SD, pois pode definir o nível de proteção contra condições ambientais adversas. Inulina, goma arábica, maltodextrinas e leite desnatado são amplamente utilizados como agentes encapsulantes para secagem de probióticos 5,17,18,19. A inulina é um frutooligossacarídeo que apresenta forte atividade prebiótica e promove a saúde intestinal19. O leite desnatado é muito eficaz na manutenção da viabilidade das células bacterianas secas e gera um pó com boas propriedades de reconstituição17.
Lactiplantibacillus paraplantarum O FT-259 é uma bactéria do ácido lático que produz bacteriocina e apresenta atividade antilisterial e características probióticas20,21. É uma bactéria Gram-positiva heterofermentativa facultativa em forma de bastonete que cresce de 15 °C a 37 °C20 e é compatível com a temperatura corporal homeostática. Este trabalho teve como objetivo apresentar todas as etapas envolvidas na produção e caracterização físico-química de um probiótico spray-dried (L. paraplantarum FT-259) e avaliar a influência dos protetores e das temperaturas de secagem.

Probióticos e prebióticos são de grande interesse para as indústrias alimentícia e farmacêutica devido aos seus benefícios para a saúde. Os probióticos são bactérias vivas que podem conferir efeitos benéficos ao bem-estar humano e animal, enquanto os prebióticos são tipos de nutrientes que alimentam as bactérias benéficas do intestino. Os probióticos em pó ganharam popularidade devido à facilidade e praticidade de sua ingestão e incorporação à dieta como suplemento alimentar. No entanto, o processo de secagem interfere na viabilidade celular, uma vez que altas temperaturas inativam bactérias probióticas. Neste contexto, este trabalho teve como objetivo apresentar todas as etapas envolvidas na produção e caracterização físico-química de um probiótico spray-dried e avaliar a influência dos protetores (leite desnatado simulado e associação inulina:maltodextrina) e das temperaturas de secagem no aumento do rendimento do pó e da viabilidade celular. Os resultados mostraram que o leite desnatado simulado promoveu maior viabilidade probiótica a 80 °C. Com esse protetor, a viabilidade do probiótico, o teor de umidade e a atividade de água (Aw) diminuem enquanto a temperatura de entrada aumenta. A viabilidade dos probióticos diminui inversamente com a temperatura de secagem. Em temperaturas próximas a 120 °C, o probiótico seco apresentou viabilidade em torno de 90%, teor de água de 4,6% p/p e Aw de 0,26; valores adequados para garantir a estabilidade do produto. Neste contexto, temperaturas de secagem por pulverização acima de 120 °C são necessárias para garantir a viabilidade e a vida útil das células microbianas na preparação em pó e a sobrevivência durante o processamento e armazenamento de alimentos.

To be defined as probiotics, microorganisms added to foods (or supplements) have to be consumed alive, be able to survive during passage in the gastrointestinal tract of the host, and reach the site of action in adequate amounts to exert beneficial effects1,2,7.
The growing interest in probiotics is due to the several benefits to human health they confer, such as the stimulation of the immune system, the reduction of serum cholesterol levels, and the enhancement of gut barrier function by acting against harmful microbes, as well as their beneficial effects in the treatment of the irritable bowel syndrome, among others2,3. In addition, several studies have demonstrated that probiotics can positively affect other parts of the human body where unbalanced microbial communities can cause infectious diseases3,4,5.
For probiotics to be therapeutically effective, the product should contain between 106-107 CFU/g of bacteria at the time of consumption6. On the other hand, the Italian Ministry of Health and Health Canada have established that the minimum level of probiotics in food should be 109 CFU/g of viable cells per day or per serving, respectively7. Considering high loads of probiotics are needed to guarantee they will have beneficial effects, it is essential to guarantee their survival during processing, shelf storage, and passage through the gastrointestinal (GI) tract. Several studies have demonstrated that microencapsulation is an effective method to improve the overall viability of probiotics8,9,10,11.
In this context, several methods have been developed for the microencapsulation of probiotics, such as spray-drying, freeze-drying, spray-chilling, emulsion, extrusion, coacervation, and, more recently, fluidized beds11,12,13,14. Microencapsulation by spray-drying (SD) is widely used in the food industry because it is a simple, fast, and reproducible process. It is easy to scale up, and it has a high production yield at low energy requirements11,12,13,14. Nonetheless, the exposure to high temperatures and low moisture content can affect the survival and viability of the probiotic cells15. Both parameters can be improved for a given strain by determining the effects of culture age and conditions to pre-adapt the culture and optimize the spray-drying conditions (inlet and outlet temperatures, atomization process) and the encapsulating composition8,14,16,17,18.
The composition of the encapsulating solution is also an important factor during SD as it can define the level of protection against adverse environmental conditions. Inulin, Arabic gum, maltodextrins, and skim milk are widely used as encapsulating agents for probiotic drying5,17,18,19. Inulin is a fructooligosaccharide that presents a strong prebiotic activity and promotes intestinal health19. Skim milk is very effective in maintaining the viability of dried bacterial cells and generates a powder with good reconstitution properties17.
Lactiplantibacillus paraplantarum FT-259 is a lactic acid bacterium that produces bacteriocin and presents antilisterial activity, besides probiotic traits20,21. It is a facultative heterofermentative rod-shaped Gram-positive bacterium that grows from 15 &#176;C to 37 &#176;C20 and is compatible with the homeostatic body temperature. This study aimed to present all the steps involved in the production and physicochemical characterization of a spray-dried probiotic (L. paraplantarum FT-259) and evaluate the influence of the protectants and drying temperatures.

Em nosso laboratório, fazemos pesquisas sobre micróbios benéficos, incluindo probióticos, que são microrganismos vivos que podem ser adicionados aos alimentos para melhorar a saúde dos consumidores, geralmente competindo com bactérias patogênicas no intestino para excluí-los e aliviar os sintomas, como diarreia ou outros distúrbios gastrointestinais. Atualmente, muitas técnicas têm sido aplicadas para garantir que os probióticos sobrevivam nos alimentos durante a vida de prateleira. E também, eles precisavam tolerar as condições adversas do trato gastrointestinal para exercer seus efeitos benéficos dentro do intestino.Estudamos muitas tecnologias que podem aumentar a sobrevivência dos probióticos, seja na alimentação ou no hospedeiro humano. Dentre as técnicas, a microencapsulação por spray drying é de grande interesse por ser simples, rápida e reprodutível, proporcionando altos rendimentos com baixa exigência energética. É desafiador, no entanto, determinar as condições certas para obter probióticos secos.E, para isso, devem ser escolhidos protetores adequados. Leite desnatado, maltodextrina, ou outro como inulina probiótica têm sido usados com diferentes cepas probióticas. A área de microencapsulação é um campo de pesquisa em evolução que tem o potencial de impactar a indústria de alimentos, fornecendo novos produtos para serem usados como ingredientes funcionais.Neste estudo, demonstramos como obter uma preparação microencapsulada de uma cultura probiótica potencial utilizando a técnica de spray drying. Também mostramos como determinar os principais parâmetros para caracterizar o pó, obtê-lo e como avaliar a taxa de sobrevivência do probiótico.
