Los factores bióticos que afectan la productividad de los cultivos son principalmente agentes patógenos y plagas. Varias plagas de insectos causan entre el 15% y el 35% de la pérdida de cultivos agrícolas y afectan las prácticas de sostenibilidad económica1. Los insectos pertenecientes a los órdenes Coleoptera, Hemiptera y Lepidoptera son los principales órdenes de plagas devastadoras. La naturaleza altamente adaptativa del medio ambiente ha beneficiado a los lepidópteros en la evolución de varios mecanismos de supervivencia. Entre los insectos lepidópteros, Helicoverpa armigera (gusano del algodón) puede alimentarse de alrededor de 180 cultivos diferentes y causar daños significativos a sus tejidos reproductivos2. A nivel mundial, la infestación por H. armigera ha provocado una pérdida de alrededor de5 mil millones de dólares. El algodón, los garbanzos, los gandules, los tomates, los girasoles y otros cultivos son huéspedes de H. armigera. Completa su ciclo de vida en diferentes partes de las plantas hospederas. Los huevos puestos por las polillas hembra eclosionan en las hojas, seguidos de su alimentación de tejidos vegetativos durante las etapas larvarias. La etapa larvaria es la más destructiva debido a su naturaleza voraz y altamente adaptable 4,5. H. armigera muestra una distribución global e invasión a nuevos territorios debido a sus notables atributos, como polifagia, excelentes capacidades migratorias, mayor fecundidad, fuerte diapausa y la aparición de resistencia a las estrategias de control de insectos existentes6.
Diversas moléculas químicas de terpenos, flavonoides, alcaloides, polifenoles, glucósidos cianogénicos y muchos otros son ampliamente utilizadas para el control de la infestación por H. armigera 7. Sin embargo, la aplicación frecuente de moléculas químicas tiene efectos adversos en el medio ambiente y la salud humana debido a la adquisición de sus residuos. Además, muestran un efecto perjudicial sobre varios depredadores de plagas, lo que resulta en un desequilibrio ecológico 8,9. Por lo tanto, existe la necesidad de investigar opciones seguras y ecológicas para las moléculas químicas de control de plagas.
Las moléculas insecticidas naturales producidas por las plantas (fitoquímicos) pueden utilizarse como una alternativa prometedora a los pesticidas químicos. Estos fitoquímicos incluyen varios metabolitos secundarios pertenecientes a las clases alcaloides, terpenoides y fenólicos 7,10. La quercetina es uno de los flavonoides (compuestos fenólicos) más abundantes presentes en diversos granos, verduras, frutas y hojas. Muestra actividad disuasoria de alimentación e insecticida contra insectos; Además, no es perjudicial para los enemigos naturales de las plagas11,12. Por lo tanto, este protocolo demuestra el ensayo de alimentación utilizando quercetina para evaluar su efecto tóxico sobre H. armigera.
Se han desarrollado varios métodos de bioensayo para evaluar el efecto de las moléculas naturales y sintéticas en los patrones de alimentación, crecimiento, desarrollo y comportamiento de un insecto13. Los métodos comúnmente utilizados incluyen el ensayo de disco foliar, el ensayo de alimentación de elección, el ensayo de alimentación por gotas, el ensayo de contacto, el ensayo de cobertura de la dieta y el ensayo de alimentación obligada13,14. Estos métodos se clasifican en función de cómo se aplican los pesticidas a los insectos. El ensayo de alimentación obligatoria es uno de los métodos más utilizados, sensibles, sencillos y adaptables para analizar los probables insecticidas y su dosis letal14. En un ensayo de alimentación obligada, la molécula de interés se mezcla con una dieta artificial. Esto proporciona consistencia y control sobre la composición de la dieta, generando resultados robustos y reproducibles. Las variables importantes que afectan a los ensayos de alimentación son la etapa de desarrollo del insecto, la elección del insecticida, los factores ambientales y el tamaño de la muestra. La duración del ensayo, el intervalo entre dos registros de datos, la frecuencia y la cantidad de dieta administrada, la salud de los insectos y la habilidad de manejo de los operadores también pueden influir en el resultado de los ensayos de alimentación14,15.
Este estudio tiene como objetivo demostrar el ensayo de alimentación obligada para evaluar el efecto de la quercetina sobre la supervivencia y aptitud física de H. armigera . La evaluación de varios parámetros, como el peso corporal de los insectos, la tasa de mortalidad y los defectos de desarrollo, proporcionará información sobre los efectos insecticidas de la quercetina. developing a feeding assay system for evaluating the insecticidal effect of phytochemicals on helicoverpa armigera

Helicoverpa armigera, un insecto lepidóptero, es una plaga polífaga con una distribución mundial. Este insecto herbívoro es una amenaza para las plantas y la productividad agrícola. En respuesta, las plantas producen varios fitoquímicos que afectan negativamente el crecimiento y la supervivencia del insecto. Este protocolo demuestra un método de ensayo de alimentación obligado para evaluar el efecto de un fitoquímico (quercetina) en el crecimiento, desarrollo y supervivencia de los insectos. The biotic factors that affect crop productivity are mainly pathogenic agents and pests. Several insect pests cause 15% to 35% of agricultural crop loss and affect economic sustainability practices1. Insects belonging to the orders Coleoptera, Hemiptera, and Lepidoptera are the major orders of devastating pests. The highly adaptive nature of the environment has benefited lepidopterans in evolving several survival mechanisms. Amongst lepidopteran insects, Helicoverpa armigera (Cotton bollworm) can feed on around 180 different crops and cause significant damage to their reproductive tissues2. Worldwide, H. armigera infestation has resulted in a loss of around $5 billion3. Cotton, chickpeas, pigeon peas, tomatoes, sunflowers, and other crops are hosts for H. armigera. It completes its lifecycle on different parts of host plants. Eggs laid by female moths get hatched on the leaves, followed by their feeding on vegetative tissues during larval stages. The larval stage is the most destructive due to its voracious and highly adaptable nature4,5. H. armigera shows a global distribution and encroachment to new territories due to its remarkable attributes, such as polyphagy, excellent migratory abilities, higher fecundity, strong diapause, and the emergence of resistance to existing insect control strategies6.
Diverse chemical molecules from terpenes, flavonoids, alkaloids, polyphenols, cyanogenic glucosides, and many others are widely used for the control of H. armigera infestation7. However, frequent application of chemical molecules imparts adverse effects on the environment and human health due to the acquisition of their residues. Also, they show a detrimental effect on various pest predators, resulting in an ecological imbalance8,9. Therefore, there is a necessity to investigate safe and eco-friendly options for chemical molecules of pest control.
Natural insecticidal molecules produced by plants (phytochemicals) can be used as a promising alternative to chemical pesticides. These phytochemicals include various secondary metabolites belonging to the classes alkaloids, terpenoids, and phenolics7,10. Quercetin is one of the most abundant flavonoids (phenolic compound) present in various grains, vegetables, fruits, and leaves. It shows feeding deterrent and insecticidal activity against insects; also, it is not harmful to natural enemies of pests11,12. Thus, this protocol demonstrates the feeding assay using quercetin to assess its toxic effect on H. armigera.
Various bioassay methods have been developed to evaluate the effect of natural and synthetic molecules on an insect&#39;s feeding, growth, development, and behavioral patterns13. Commonly used methods include the leaf disk assay, choice feeding assay, droplet feeding assay, contact assay, diet covering assay, and obligate feeding assay13,14. These methods are classified based on how pesticides are applied to insects. The obligate feeding assay is one of the most commonly used, sensitive, simple, and adaptable methods to test probable insecticides and their lethal dose14. In an obligate feeding assay, the molecule of interest is mixed with an artificial diet. This provides consistency and control over the diet composition, generating robust and reproducible results. Important variables affecting feeding assays are the developmental stage of the insect, choice of insecticide, environmental factors, and sample size. The duration of the assay, interval between two data recordings, frequency and amount of diet fed, health of insects, and handling skill of operators can also influence the outcome of feeding assays14,15.
This study aims to demonstrate the obligate feeding assay to evaluate the effect of quercetin on H. armigera survival and fitness. Assessment of various parameters, such as insect body weight, mortality rate, and developmental defects, will provide insights into the insecticidal effects of quercetin. Meanwhile, measuring nutritional parameters, including the efficiency of conversion of ingested food (ECI), efficiency of conversion of digested food (ECD), and approximate digestibility (AD), will highlight the antifeedant attributes of quercetin.

This study is conducted to develop a simple and cost-effective feeding assay method for evaluating the effect of various natural molecules, including phytochemicals for their antifeedant and insecticidal properties against thieving insects, and identified potential molecules from these assay can be evaluated in field condition for pest management. Multiple assays have been developed to evaluate the effect of synthetic and natural molecules on insect feeding, growth, development, and behavior, further assays have been designed, such as dip case tool, diet covering assay, contact assay, droplet feeding assay, choice feeding assay, and lip dish test for insecticide evaluation, Maintaining uniform experimental conditions, addressing issues, like infection and cannibalism, ensuring even phytochemical distribution in larval diet, tracking, feeding behavior accurately, and determining optimal phytochemical concentrations post challenges, batch variation, and experimental design complexities must be managed. Additionally, transitioning from the lab assist to the field application, considering formulation and scalability is demanding.In the obligatory feeding assay, a large number of molecules can be tested simultaneously with a large number of insect sample sizes. The advantage of this feeding assay is that the test compound is mixed with an artificial diet, thus we have control over diet composition, which provides robust and reproducible results.
