Ons onderzoek is gericht op het ontwikkelen van nieuwe hulpmiddelen om afzonderlijke moleculen te detecteren door oppervlakte-versterkte Raman-verstrooiing of bron. Dit is de enige techniek die een chemische vingerafdruk van een molecuul biedt en gevoelig genoeg is om afzonderlijke moleculen te detecteren. Op deze manier kan gedetailleerde mechanistische informatie over chemische reacties worden verkregen.
DNA origami nanostructuren zijn gebruikt om zowel plasmonische nanodeeltjes als doelmoleculen nauwkeurig te positioneren. En dit is nodig omdat de verbeterde Raman-verstrooiing afkomstig is van een klein nanometrisch volume tussen de nanodeeltjes die we hotspots noemen. En we hebben nu nieuwe plasmonische DNA origami nanoantenna gemaakt, precies voor dit doel.
De belangrijkste uitdaging is om doelmoleculen in dergelijke hotspots tussen twee nanodeeltjes te plaatsen en Raman-gegevens te verzamelen van precies één nanoantenna-structuur. Om grote hoeveelheden gegevens en een efficiënte correlatie tussen atoomkrachtmicroscopie te verzamelen, moet een Raman-spectroscopie worden uitgevoerd. De plasmonische DNA origami nanoantenna's zorgen voor een reproduceerbare productie van een groot aantal plasmonische dimeren waarbij het doelmolecuul precies tussen de nanodeeltjes in de hotspot is geplaatst.
En door een correlatie van de AFM- en Raman-gegevens kunnen we er nu voor zorgen dat er slechts één molecuul wordt gedetecteerd. Nu kunnen we afzonderlijke moleculen zoals kleurstofmoleculen of eiwitten in realtime volgen, en hun gedrag in de hotspots en hun reacties op chemische veranderingen in de omgeving. Zo werd onlangs de verandering van de spintoestand van afzonderlijke menselijke moleculen gemonitord.
In de toekomst willen we chemische reacties op moleculair niveau volgen en hun reactiemechanismen bestuderen. Daarnaast kunnen we deze technologie gebruiken om medisch relevante biomoleculen met een zeer hoge gevoeligheid te detecteren.
