We beschrijven een protocol voor het genereren van menselijke podocyten door episomale herprogrammering van dermale fibroblasten in door de mens geïnduceerde pluripotente stamcellen. Deze cellen lijken veel beter op in vivo podocyten dan vereeuwigde podocyten in termen van podocytenvoedselprocessen en expressie van podocytspecifieke marker. Podocyten zijn terminaal gedifferentieerde cellen.
Daarom vermenigvuldigen deze cellen zich niet langer en is de primaire podocytenanalyse in celkweek beperkt. Hoewel het probleem van het beperkte celaantal werd overwonnen door voorwaardelijk vereeuwigde podocyten te gebruiken, vertonen deze podocyten een gededifferentieerd fenotype en gedrag, dat hun gebruik in fundamenteel onderzoek in twijfel trekt. Met behulp van geïnduceerde pluripotente stamcellen kunnen podocyten in vitro worden gegenereerd in een bijna onbeperkt celaantal met typische podocytenmorfologie, inclusief verschillende voedselprocessen en expressie van podocytspecifieke marker.
Bij het gebruik van de patiëntspecifieke stamcellen om te differentiëren in podocyten, is het mogelijk om de ziektespecifieke veranderingen van de cel ex vivo te bestuderen. In het geval dat de eerste huidbiopsie werd genomen van een patiënt met een genetische podocytenziekte. Podocyten gegenereerd met ons protocol zijn gepersonaliseerde zieke cellen die de mutatie van de patiënt dragen.
Deze cellen vertegenwoordigen dus een verbeterd ex vivo model om podocytenziekten en potentiële therapeutische stoffen te bestuderen in een geïndividualiseerde benadering.
