Wir beschreiben ein Protokoll zur Generierung humaner Podozyten durch episomale Reprogrammierung von dermalen Fibroblasten in human-induzierte pluripotente Stammzellen. Diese Zellen ähneln in vivo Podozyten viel besser als immortalisierten Podozyten in Bezug auf Podozyten-Nahrungsprozesse und die Expression von Podozyten-spezifischen Markern. Podozyten sind terminal differenzierte Zellen.
Daher vermehren sich diese Zellen nicht mehr und die primäre Podozytenanalyse in der Zellkultur ist eingeschränkt. Obwohl das Problem der begrenzten Zellzahl durch die Verwendung von bedingt immortalisierten Podozyten überwunden werden konnte, weisen diese Podozyten einen dedifferenzierten Phänotyp und ein dedifferenziertes Verhalten auf, was ihre Verwendung in der Grundlagenforschung in Frage stellt. Mit Hilfe von induzierten pluripotenten Stammzellen können Podozyten in vitro in einer nahezu unbegrenzten Zellzahl mit typischer Podozytenmorphologie einschließlich unterschiedlicher Nahrungsprozesse und der Expression von Podozyten-spezifischen Markern erzeugt werden.
Durch die Verwendung der patientenspezifischen Stammzellen zur Differenzierung in Podozyten ist es möglich, die krankheitsspezifischen Veränderungen der Zelle ex vivo zu untersuchen. In diesem Fall wurde die erste Hautbiopsie von einem Patienten mit einer genetisch bedingten Podozytenerkrankung entnommen. Podozyten, die mit unserem Protokoll erzeugt werden, sind personalisierte kranke Zellen, die die Mutation des Patienten tragen.
Somit stellen diese Zellen ein verbessertes ex vivo Modell dar, um Podozytenerkrankungen und potentielle therapeutische Substanzen in einem individualisierten Ansatz zu untersuchen.
