Vi beskriver en protokoll for generering av humane podocytter ved episomal omprogrammering av dermale fibroblaster til menneskeskapte pluripotente stamceller. Disse cellene ligner in vivo podocytter mye bedre enn immortaliserte podocytter når det gjelder podocytmatprosesser og ekspresjon av podocyttspesifikke markører. Podocytter er terminalt differensierte celler.
Derfor prolifererer disse cellene ikke lenger, og primær podocyttanalyse i cellekultur er begrenset. Selv om problemet med begrenset cellenummer ble overvunnet ved bruk av betinget udødeliggjorte podocytter, presenterer disse podocyttene en dedifferensiert fenotype og oppførsel, som stiller spørsmål ved deres bruk i grunnforskning. Ved hjelp av induserte pluripotente stamceller kan podocytter genereres in vitro i et nesten ubegrenset celletall med typisk podocytmorfologi, inkludert distinkte matprosesser og ekspresjon av podocyttspesifikk markør.
Ved bruk av pasientspesifikke stamceller for å differensiere til podocytter, er det mulig å studere cellens sykdomsspesifikke endringer ex vivo. I tilfelle ble den første hudbiopsien tatt fra en pasient med en genetisk podocyttsykdom. Podocytter generert med vår protokoll er personlige syke celler som bærer pasientens mutasjon.
Dermed representerer disse cellene en forbedret ex vivo-modell for å studere podocyttsykdommer og potensielle terapeutiske stoffer i en individualisert tilnærming.
