Además, utilizamos modelos celulares y animales para probar posibles tratamientos contra la enfermedad de Parkinson. En este artículo se propone un protocolo para el estudio de la morfología mitocondrial in situ como indicador de su salud basado en inmunotinción y análisis de imagen en cortes de tejido embebidos en parafina.
La morfología mitocondrial ha sido ampliamente estudiada en la enfermedad de Parkinson y otras enfermedades in vitro. Sin embargo, aún faltan protocolos para estudiar la morfología mitocondrial in vivo. Este protocolo proporciona un método robusto para teñir mitocondrias en tejido cerebral, y explica en detalle cómo realizar el análisis morfológico.
Este estudio muestra que la inmunotinción combinada con el análisis de imágenes es un método fiable para comprender la morfología mitocondrial. De hecho, permite cuantificar el número de mitocondrias así como otros parámetros morfológicos, como la relación de aspecto tanto en cultivo celular como en tejido. Es importante destacar que, dado que este método se basa en la inmunofluorescencia, es posible relacionar las morfologías con poblaciones celulares específicas.
Aquí presentamos un protocolo para el estudio de la morfología mitocondrial en la enfermedad de Parkinson, pero este protocolo se puede aplicar a cualquier modelo de enfermedad in vivo. Además, puede ayudar a detectar compuestos de plomo para el tratamiento no solo de la enfermedad de Parkinson, sino también de cualquier otra enfermedad caracterizada por la desregulación mitocondrial.
