We zijn geïnteresseerd in overspraak tussen samentrekkende spieren en de adipocyten die zich vormen tussen spiervezels bij letsel en ziekte die intramusculair vetweefsel of IMAT wordt genoemd. Genetische manipulatie in cellen bij muizen is een krachtig hulpmiddel voor het mechanistisch ontleden van overspraak van vetspieren. Analyse van signalering via transcriptomics en proteomics identificeert mediatoren, en vervolgens kan op biomateriaal gebaseerde medicijnafgifte ingrijpen in deze routes.
Een van de belangrijkste uitdagingen is het isoleren en kwantificeren van IMAT-adipocyten. Vooral in kleine diermodellen wordt het IMAT-signaal weggespoeld door analyse van de hele spier, zoals transcriptomics of lipidomics, en het kwantificeren van IMAT door niet-invasieve beeldvorming lijdt aan een vergelijkbare uitwassing, aangezien de meeste voxels een mengsel van spier en IMAT bevatten. We hebben deze techniek die in dit artikel wordt beschreven onlangs gebruikt om aan te tonen dat IMAT de spiercontractiliteit direct schaadt.
Met nauwkeurige metingen van IMAT-afzetting toonden we aan dat het niet alleen contractiel materiaal vervangt, maar ook een contractie in de resterende droge spier belemmert. Dit protocol pakt uitdagingen aan bij het kwantificeren van IMAT. De huidige standaard voor het meten van IMAT bij kleine dieren is histologie met één sectie, wat geen uitgebreide of niet-invasieve beeldvorming is, wat duur is en een lage resolutie heeft.
We zijn van mening dat dit ons begrip van IMAT-spieroverspraak heeft beperkt. Deze techniek biedt een uitgebreide en goedkope methode voor het beoordelen van IMAT door middel van kwalitatieve visualisatie en kwantificering op meerdere schalen. We hopen dat de eenvoud van deze techniek meer onderzoekers zal aanmoedigen om IMAT in hun diermodellen te meten.
Deze techniek zal ook een nauwkeuriger hulpmiddel bieden voor de onderzoekers die geïnteresseerd zijn in IMAT om subtielere relaties tussen IMAT-adipocyten en andere cellen te ontdekken.
