Immunfluorescensanalyse av γH2AX og 53BP1 foci er en egnet metode for å vurdere genomisk skade i interfasekjerner i et cellesystem. DNA-skade induseres kontinuerlig av midler som kan være endogene, for eksempel ROS generert av cellulær oksidativ metabolisme, eller eksogene, både kjemikalier og fysisk1. Blant de mest skadelige lesjonene spiller dobbeltstrengbrudd (DSB) en grunnleggende rolle i å bidra til genomisk ustabilitet, siden de forårsaker kromosomavvik som igjen kan starte karsinogeneseprosessen. Dermed er cellene utstyrt med komplekse og effektive mekanismer for DSBs reparasjon2.
Når en DSB oppstår, utløser cellen DNA-skaderesponsen (DDR) der ATM- eller ATR-kinaser sammen med MRE11 / RAD50 / NBS1-komplekset rekrutteres for å aktivere andre proteiner som bremser eller stopper cellesyklusen3. Et viktig mål for disse kinasene er histon H2AX, som fosforyleres på Ser-139 i noen få megabaser fra DSB-ene (nemlig γH2AX), og dermed tillater rekruttering av flere reparasjonsfaktorer som blant annet BRCA1 og p53 bindingsprotein 1 (53BP1)3. Senere utløses en vei mellom homolog rekombinasjon (HR), ikke-homolog endesammenføyning (NHEJ) eller enkeltstrenget glødning (SSA) for å reparere DSB 4,5. Derfor er 53BP1 involvert i å diktere valget mellom HR eller NHEJ, hovedsakelig fremme aktiveringen av NHEJ i stedet for HR6. Videre kan både den fosforylerte formen av H2AX histon og 53BP1 danne foci på stedene til DSB. Da disse fociene vedvarer til integriteten til dobbeltstrengen er gjenopprettet, anses det å vurdere utseendet / forsvinningen av γH2AX eller 53BP1 foci innen et tidsintervall som en nyttig metode for å evaluere forekomsten og reparasjonen av DSB i et cellesystem 6,7. Dobbeltstrengbrudd (DSB) er en av de mest alvorlige lesjonene som kan oppstå i cellekjerner, og hvis de ikke repareres, kan de føre til alvorlige utfall, inkludert kreft. Cellen er derfor utstyrt med komplekse mekanismer for å reparere DSB, og disse veiene involverer histon H2AX i sin fosforylerte form ved Ser-139 (nemlig γH2AX) og p53 bindende protein 1 (53BP1). Data oppnådd ved fluorescensmikroskopanalyse av perifere lymfocytter tillater oss å evaluere tre hovedaspekter: effektiviteten av bleomycinbehandling ved å øke antallet γH2AX og 53BP1 foci (og dermed DSB) på grunn av dens mutagene effekt, i hvilken grad begge foci samlokaliseres på stedet for DSB, og tidsforløpet av γH2AX og 53BP1 foci for å avgrense reparasjonskinetikken til bleomycininduserte DSB. Målet med dette manuskriptet var å evaluere egnetheten av samtidig kvantifisering av Gamma H2AX og 53BP1 foci for å vurdere genomisk skade indusert i humane perifere lymfocytter av bleomycin. Flere faktorer kan forårsake dobbeltstrengbrudd. Cellene er dermed utstyrt med flere reparasjonsmekanismer, som involverer både Gamma H2AX og 53 Binding Protein 1, som kan samlokaliseres nær dobbeltstrengbruddet.Etter å ha samlet hele blodprøver, tilsett 300 mikroliter prøve til et rør som inneholder 4, 7 ml komplett medium. Deretter tilsettes fem mikrogram per milliliter bleomycinsulfat sluttkonsentrasjon. Definer en negativ kontroll for hvert eksempel.Sett røret i en termostat ved 37 grader Celsius i to timer. Humane lymfocytter uten Gamma H2AX og 53BP1 foci er vist i A, i B og i C er humane lymfocytter med forskjellige mengder Gamma H2AX og 53BP1 foci vist.Det observeres et lavt nivå av foci samlokalisering. Som forventet observeres en svært høyere frekvens av både Gamma H2AX og 53BP1 foci mellom ubehandlede ubehandlede celler. Også en stor forskjell observeres i antall foci av de to markørene.Tidsforløpet av Gamma H2AX og 53BP1 foci over tid viste en annen oppførsel, selv om de bidrar til samme funksjon. Gamma H2AX og 53BP1 foci analyse har evnen til å evaluere den cellulære responsen i enten eksponering eller patologiske sammenhenger. Imidlertid kan Gamma H2AX og 53BP1 dobbel immunfluorescens føre til underestimering av den sanne frekvensen av hendelsen.
