Ons onderzoek onderzoekt hoe mechanische signalen het lot van endotheelcellen tijdens de ontwikkeling instrueren, waarvoor we door de mens geïnduceerde pluripotente stamceldifferentiatie combineren met fluïdisch gemedieerde mechanische stimulatie. Endotheelcellen worden constant blootgesteld aan de beweging van bloed, dus belangrijke signalen worden gemist in statische kweek, wat dit de grootste uitdaging maakt bij het bestuderen van endotheelcelbiologie met behulp van traditionele kweeksystemen. Mechanische signalen activeren in feite veel cruciale signaalroutes die over het hoofd kunnen worden gezien in cellen die in statische omstandigheden worden gekweekt.
Ons protocol biedt een eenvoudige, stapsgewijze handleiding voor het differentiëren en mechanisch stimuleren van geïnduceerde pluripotente endotheelcellen uit stamcellen. Dit zal veel laboratoria helpen bij het adopteren van een thermische kweek om gemakkelijk vragen te beantwoorden over het gedrag van endotheelcellen terwijl ze onder stroom zijn.
