Nossa pesquisa visa entender como o ciclo celular é regulado em ovócitos de mamíferos e em embriões pré-implantacionais. Um aspecto chave de nossa pesquisa envolve o exame da capacidade de reparo e resposta a danos ao DNA em diferentes estados de maturação em ovócitos e também em embriões pré-implantação. Estamos particularmente interessados em entender como os pontos de verificação respondem e quais mecanismos de reparo estabelecem após possíveis danos ao DNA.
Nosso laboratório foi o primeiro a examinar mecanisticamente a resposta a danos ao DNA em ovócitos de mamíferos. Nós mostramos a ausência do ponto de verificação de danos ao DNA pró-fase G2 em ovócitos de mamíferos. E observamos a importância do ponto de verificação da montagem do fuso no estabelecimento da parada metafásica após o dano.
Usando este protocolo, é possível detectar quebras de fita dupla de DNA com precisão e determinar se os ovócitos têm danos no DNA e se os danos ao DNA estão sendo reparados. Este protocolo envolve o uso de imunofluorescência que permite identificar os locais exatos de dano ao DNA.
