Transplantation er ofte den eneste effektive behandling for organsygdom i slutstadiet. For at forstå de komplekse mekanismer, der er forbundet med skade ved levertransplantation, er der udviklet robuste transplantationsmodeller for små dyr for at rekapitulere fysiologien og patofysiologien af allograftskade. Der skal udvikles innovative strategier for i dybden at undersøge de mekanistiske veje, der er involveret i vævsskader, der observeres ved transplantation, og for at udvide donorpuljen.
Brugen af mus til at studere transplantation er afgørende, da det giver mulighed for brug af genredigerede og transgene muselinjer til at studere rollen af specifikke molekylære veje på patofysiologien af allograftskade. Der er en række tekniske udfordringer med muselevertransplantation, herunder den lille diameter af den vaskulære anastomose og den efterfølgende risiko for trombose, der fører til allograftsvigt. Mens mange teknologier såsom ultralyd angiografi er blevet anvendt, mikrocomputer tomografi har fordele, herunder evnen til at opnå høj opløsning billeder af vaskulær anastomose i hele maven.
Disse undersøgelser kan udføres i længderetningen for at evaluere vaskulær patency og overflod af leveren over tid. Der er mange udfordringer ved at udføre nøjagtige og klare mikro CT angiografi undersøgelser hos gnavere. Vi har fundet ud af, at brugen af hjertegating og justering af isoflurankoncentrationer for at reducere respirationsfrekvensen giver de klareste billeder.
Vi har også fundet ud af, at brug af gnaverspecifik kontrasttiming til specifikke faser også har forbedret visualiseringen.
