Transplantatie is vaak de enige effectieve therapie voor orgaanziekte in het eindstadium. Om de complexe mechanismen te begrijpen die inherent zijn aan letsel door transplantatie in de lever, zijn robuuste transplantatiemodellen voor kleine dieren ontwikkeld om de fysiologie en pathofysiologie van allotransplantaatletsel te recapituleren. Er zullen innovatieve strategieën moeten worden ontwikkeld om de mechanistische routes die betrokken zijn bij weefselbeschadiging die bij transplantatie wordt waargenomen, diepgaand te onderzoeken en om de donorpool uit te breiden.
Het gebruik van muizen om transplantatie te bestuderen is van vitaal belang omdat het het gebruik van genbewerkte en transgene muislijnen mogelijk maakt om de rol van specifieke moleculaire routes op de pathofysiologie van allotransplantaatletsel te bestuderen. Er zijn een aantal technische uitdagingen bij levertransplantatie bij muizen, waaronder de kleine diameter van de vasculaire anastomose en het daaropvolgende risico op trombose die leidt tot het falen van het transplantaat. Hoewel er veel technologieën zijn gebruikt, zoals ultrasone angiografie, heeft microcomputertomografie voordelen, waaronder de mogelijkheid om beelden met een hoge resolutie van de vasculaire anastomose in de hele buik te verkrijgen.
Deze onderzoeken kunnen longitudinaal worden uitgevoerd om de doorgankelijkheid van de vasculaire en de overvloed van de lever in de loop van de tijd te evalueren. Er zijn veel uitdagingen bij het uitvoeren van nauwkeurige en duidelijke micro-CT-angiografiestudies bij knaagdieren. We hebben ontdekt dat het gebruik van cardiale gating en de aanpassing van isofluraanconcentraties om de ademhalingsfrequentie te verlagen, de duidelijkste beelden opleveren.
We hebben ook ontdekt dat het gebruik van knaagdierspecifieke contrasttiming voor specifieke fasen ook de visualisatie heeft verbeterd.
