Transplantasjon er ofte den eneste effektive terapi for sluttstadiet organsykdom. For å forstå de komplekse mekanismene som er forbundet med skade ved levertransplantasjon, har robuste små dyrtransplantasjonsmodeller blitt utviklet for å rekapitulere fysiologien og patofysiologien til allograftskade. Innovative strategier må utvikles for å dypt forhøre de mekanistiske veiene som er involvert i vevsskade observert ved transplantasjon, og for å utvide donorbassenget.
Bruken av mus for å studere transplantasjon er viktig, da det muliggjør bruk av genredigerte og transgene muselinjer for å studere rollen som spesifikke molekylære veier på patofysiologien av allograftskade. Det er en rekke tekniske utfordringer ved levertransplantasjon fra mus, inkludert den lille diameteren av vaskulær anastomose og den påfølgende risikoen for trombose som fører til allotransplantatsvikt. Mens mange teknologier som ultralydangiografi har blitt ansatt, har mikrocomputertomografi fordeler, inkludert evnen til å oppnå høyoppløselige bilder av vaskulær anastomose i hele magen.
Disse studiene kan utføres i lengderetningen for å evaluere vaskulær patency og overflod av leveren over tid. Det er mange utfordringer med å utføre nøyaktige og klare mikro CT-angiografistudier hos gnagere. Vi har funnet at bruk av hjertegating og justering av isofluran for å redusere respirasjonsfrekvensen gir de klareste bildene.
Vi har også funnet ut at bruk av gnagerspesifikk kontrasttiming for spesifikke faser også har forbedret visualisering.
