Transplantation är ofta den enda effektiva behandlingen för organsjukdom i slutstadiet. För att förstå de komplexa mekanismer som är inneboende i skador vid levertransplantation har robusta smådjurstransplantationsmodeller utvecklats för att rekapitulera fysiologin och patofysiologin vid transplantatskada. Innovativa strategier kommer att behöva utvecklas för att på djupet undersöka de mekanistiska vägar som är involverade i vävnadsskador som observeras vid transplantation och för att utöka donatorpoolen.
Användningen av möss för att studera transplantation är avgörande eftersom det möjliggör användning av genredigerade och transgena muslinjer för att studera rollen av specifika molekylära vägar på patofysiologin vid allograftskada. Det finns ett antal tekniska utmaningar med levertransplantation på möss, inklusive den lilla diametern på den vaskulära anastomosen och den efterföljande risken för trombos som leder till allograftfel. Även om många tekniker som ultraljudsangiografi har använts, har mikrodatortomografi fördelar, inklusive förmågan att få högupplösta bilder av den vaskulära anastomosen i hela buken.
Dessa studier kan utföras longitudinellt för att utvärdera vaskulär patency och leverns överflöd över tid. Det finns många utmaningar med att utföra noggranna och tydliga mikro-CT-angiografistudier på gnagare. Vi har funnit att användningen av hjärtgrind och justering av isoflurankoncentrationer för att minska andningsfrekvensen ger de tydligaste bilderna.
Vi har också funnit att användningen av gnagarspecifik kontrasttiming för specifika faser också har förbättrat visualiseringen.
