Vores forskningsprogram fokuserer på strukturelle og funktionelle aspekter af mitokondrier, især da de vedrører aldringsrelateret dysfunktion. Til dette formål bruger vi en bred vifte af biofysiske, biokemiske og strukturelle teknikker til at udforske de grundlæggende mekanismer for mitokondriel aldring og til at udvikle terapeutiske interventioner for at bevare og genoprette mitokondriel sundhed. Mitokondriel funktion og struktur er uløseligt forbundet.
Standard lysmikroskopiteknikker er velegnede til måling af mitokondriefunktioner i stor skala som grundlæggende morfologi og netværksstrukturer. Analyse af mitokondriel ultrastruktur eller funktioner, der kræver højere opløsning end ved traditionel optisk mikroskopi, udføres typisk ved hjælp af elektronmikroskopi eller tomografi på faste prøver. Superopløsningsmikroskopi er en fluorescensbaseret billeddannelsesteknik, der måler funktioner ud over diffraktionsgrænsen.
For mitokondrier omfatter dette målinger af proteinfordeling i nanoskala og cristae-arkitektur. Den stead imaging protokol, der er beskrevet her, giver forskere mulighed for at måle den detaljerede struktur af den indre membran i levende celler. Live cellebilleddannelse giver os mulighed for at observere og kvantitativt analysere komplekse træk ved cristae under fysiologisk relevante forhold uden behov for prøvefiksering.
Dette vil give forskere ny indsigt i de dynamiske ændringer, der forekommer i ultrastrukturelle træk og deres tidsmæssige reaktioner på stressorer og farmakologiske forbindelser.
