Ons onderzoeksprogramma richt zich op structurele en functionele aspecten van mitochondriën, met name als ze betrekking hebben op verouderingsgerelateerde disfunctie. Daartoe gebruiken we een breed scala aan biofysische, biochemische en structurele technieken om de basismechanismen van mitochondriale veroudering te onderzoeken en therapeutische interventies te ontwikkelen om de mitochondriale gezondheid te behouden en te herstellen. Mitochondriale functie en structuur zijn onlosmakelijk met elkaar verbonden.
Standaard lichtmicroscopietechnieken zijn geschikt voor het meten van grootschalige mitochondriale kenmerken zoals basismorfologie en netwerkstructuren. Het analyseren van de mitochondriale ultrastructuur of kenmerken die een hogere resolutie vereisen dan traditionele optische microscopie, wordt meestal gedaan met behulp van elektronenmicroscopie of tomografie op vaste monsters. Superresolutiemicroscopie is een op fluorescentie gebaseerde beeldvormingstechniek die kenmerken meet die verder gaan dan de diffractielimiet.
Voor mitochondriën omvat dit metingen van eiwitverdeling op nanoschaal en cristae-architectuur. Het hier beschreven stead imaging-protocol stelt onderzoekers in staat om de gedetailleerde structuur van het binnenmembraan in levende cellen te meten. Beeldvorming van levende cellen stelt ons in staat om complexe kenmerken van cristae te observeren en kwantitatief te analyseren onder fysiologisch relevante omstandigheden zonder dat monsterfixatie nodig is.
Dit zal onderzoekers nieuwe inzichten geven in de dynamische veranderingen die optreden in ultrastructurele kenmerken en hun temporele reacties op stressoren en farmacologische verbindingen.
