Unser Forschungsprogramm konzentriert sich auf strukturelle und funktionelle Aspekte der Mitochondrien, insbesondere im Zusammenhang mit altersbedingten Dysfunktionen. Zu diesem Zweck verwenden wir ein breites Spektrum an biophysikalischen, biochemischen und strukturellen Techniken, um die grundlegenden Mechanismen des mitochondrialen Alterns zu erforschen und therapeutische Interventionen zur Erhaltung und Wiederherstellung der mitochondrialen Gesundheit zu entwickeln. Mitochondriale Funktion und Struktur sind untrennbar miteinander verbunden.
Standard-Lichtmikroskopie-Techniken eignen sich für die Messung großräumiger mitochondrialer Merkmale wie grundlegende Morphologie und Netzwerkstrukturen. Die Analyse der mitochondrialen Ultrastruktur oder von Merkmalen, die eine höhere Auflösung als die herkömmliche optische Mikroskopie erfordern, erfolgt in der Regel mit Elektronenmikroskopie oder Tomographie an fixierten Proben. Die hochauflösende Mikroskopie ist ein fluoreszenzbasiertes Bildgebungsverfahren, das Merkmale jenseits der Beugungsgrenze misst.
Für Mitochondrien umfasst dies Messungen der nanoskaligen Proteinverteilung und der Cristae-Architektur. Das hier beschriebene Stead-Imaging-Protokoll ermöglicht es den Forschern, die detaillierte Struktur der inneren Membran in lebenden Zellen zu messen. Die Bildgebung lebender Zellen ermöglicht es uns, komplexe Merkmale von Cristae unter physiologisch relevanten Bedingungen zu beobachten und quantitativ zu analysieren, ohne dass eine Probenfixierung erforderlich ist.
Dies wird den Forschern neue Einblicke in die dynamischen Veränderungen geben, die in ultrastrukturellen Merkmalen auftreten, und ihre zeitlichen Reaktionen auf Stressoren und pharmakologische Verbindungen.
