Vårt forskningsprogram fokuserer på strukturelle og funksjonelle aspekter av mitokondrier, særlig når de relaterer seg til aldringsrelatert dysfunksjon. For dette formål bruker vi et bredt spekter av biofysiske, biokjemiske og strukturelle teknikker for å utforske de grunnleggende mekanismene for mitokondriell aldring og å utvikle terapeutiske inngrep for å bevare og gjenopprette mitokondriell helse. Mitokondriell funksjon og struktur er uløselig relatert.
Standard lysmikroskopiteknikker er egnet for måling av storskala mitokondrielle egenskaper som grunnleggende morfologi og nettverksstrukturer. Analyse av mitokondriell ultrastruktur eller funksjoner som krever høyere oppløsning enn tradisjonell optisk mikroskopi gir, gjøres vanligvis ved hjelp av elektronmikroskopi eller tomografi på faste prøver. Superoppløsningsmikroskopi er en fluorescensbasert bildebehandlingsteknikk som måler funksjoner utover diffraksjonsgrensen.
For mitokondrier inkluderer dette målinger av nanoskalaproteinfordeling og cristae-arkitektur. Stead imaging-protokollen beskrevet her gjør det mulig for forskere å måle den detaljerte strukturen til den indre membranen i levende celler. Levende celleavbildning lar oss observere og kvantitativt analysere komplekse egenskaper av cristae under fysiologisk relevante forhold uten behov for prøvefiksering.
Dette vil gi forskere ny innsikt i de dynamiske endringene som forekommer i ultrastrukturelle egenskaper og deres tidsmessige responser på stressorer og farmakologiske forbindelser.
