Nuestro programa de investigación se centra en los aspectos estructurales y funcionales de las mitocondrias, particularmente en lo que se refiere a la disfunción relacionada con el envejecimiento. Con este fin, utilizamos una amplia gama de técnicas biofísicas, bioquímicas y estructurales para explorar los mecanismos básicos del envejecimiento mitocondrial y desarrollar intervenciones terapéuticas para preservar y restaurar la salud mitocondrial. La función y la estructura mitocondrial están inextricablemente relacionadas.
Las técnicas estándar de microscopía óptica son adecuadas para medir características mitocondriales a gran escala, como la morfología básica y las estructuras de red. El análisis de la ultraestructura mitocondrial o de las características que requieren una resolución más alta que la que ofrece la microscopía óptica tradicional se realiza normalmente mediante microscopía electrónica o tomografía en muestras fijas. La microscopía de superresolución es una técnica de imagen basada en fluorescencia que mide características más allá del límite de difracción.
En el caso de las mitocondrias, esto incluye mediciones de la distribución de proteínas a escala nanométrica y la arquitectura de las crestas. El protocolo de imágenes stead descrito aquí permite a los investigadores medir la estructura detallada de la membrana interna en las células vivas. Las imágenes de células vivas nos permiten observar y analizar cuantitativamente características complejas de las crestas en condiciones fisiológicamente relevantes sin necesidad de fijar la muestra.
Esto proporcionará a los investigadores nuevos conocimientos sobre los cambios dinámicos que se producen en las características ultraestructurales y sus respuestas temporales a los factores estresantes y a los compuestos farmacológicos.
