Vårt forskningsprogram fokuserar på strukturella och funktionella aspekter av mitokondrier, särskilt när de relaterar till åldersrelaterad dysfunktion. För detta ändamål använder vi ett brett spektrum av biofysiska, biokemiska och strukturella tekniker för att utforska de grundläggande mekanismerna för mitokondriellt åldrande och för att utveckla terapeutiska interventioner för att bevara och återställa mitokondriell hälsa. Mitokondriell funktion och struktur är oupplösligt relaterade.
Standardljusmikroskopitekniker är lämpliga för att mäta storskaliga mitokondriella egenskaper som grundläggande morfologi och nätverksstrukturer. Analys av mitokondriell ultrastruktur eller egenskaper som kräver högre upplösning än vad som erbjuds av traditionell optisk mikroskopi görs vanligtvis med hjälp av elektronmikroskopi eller tomografi på fasta prover. Superupplösningsmikroskopi är en fluorescensbaserad avbildningsteknik som mäter egenskaper bortom diffraktionsgränsen.
För mitokondrier inkluderar detta mätningar av proteindistribution i nanoskala och cristae-arkitektur. Det här "stead imaging"-protokollet som beskrivs här gör det möjligt för forskare att mäta den detaljerade strukturen hos det inre membranet i levande celler. Avbildning av levande celler gör det möjligt för oss att observera och kvantitativt analysera komplexa egenskaper hos Cristae under fysiologiskt relevanta förhållanden utan behov av provfixering.
Detta kommer att ge forskare nya insikter om de dynamiska förändringar som sker i ultrastrukturella egenskaper och deras tidsmässiga svar på stressorer och farmakologiska föreningar.
