Ons doel is om een nauwkeurig en gelokaliseerd alkalisch letsel in het hoornvlies en limbale gebied in een muismodel te induceren. We ontwikkelden een nieuwe punch-trephine-methode en bestudeerden vervolgens hoe effectief deze methode is bij het creëren van een cornea- en limbale stamceldeficiëntie. Geavanceerde technieken op het gebied van behandeling van hoornvliestroebelheid en regeneratie van het hoornvlies, waaronder limbale stamceltransplantatie, exosoomtransplantatie en mitochondriëntransplantatie, zijn gebaseerd op onderzoeken die de reacties van het hoornvlies op verschillende verwondingen en behandelingen hebben onderzocht.
Daarom is het van cruciaal belang om een standaard, nauwkeurig en betrouwbaar letselmodel onder onderzoekers te hebben. Er zijn verschillende technieken en diermodellen gebruikt om verschillende hoornvliesletsels te induceren en verschillende aspecten van het genezingsproces van het hoornvlies te onderzoeken. Het alkalibrandmodel is een gerenommeerd verwondingsmodel, dat wordt uitgevoerd door natriumhydroxide rechtstreeks op het hoornvliesoppervlak aan te brengen of door plat filtreerpapier te gebruiken.
We ontwikkelen een nieuwe punch-trephine-methode om nauwkeurig en betrouwbaar een muismodel van limbale stamceldeficiëntie te creëren, de beperkingen van eerdere technieken te overwinnen en het risico op onvoldoende letsel aan het limbale gebied of extra letsel aan de fornix en het bindvlies te minimaliseren. Deze techniek heeft verschillende voordelen. Effectieve chemische verwonding van het gehele hoornvliesoppervlak en limbus bij muizen, gelokaliseerde en goed omschreven verwonding van het hoornvlies, toepassing van een vloeistof gedurende een vooraf bepaalde duur, inductie van verschillende hoornvliesletselgroottes met geschikte biopsieponsen.
Het is ook haalbaar voor modellen met ratten- en konijnenverwondingen.
