Vårt mål er å indusere en presis og lokalisert alkalisk skade i hornhinnen og limbalområdet i en musemodell. Vi utvikler en ny punch-trephine metode, og studerte deretter hvor effektiv denne metoden er i å skape en hornhinne og limbal stamcellemangel. Avanserte teknikker innen hornhinneopasitetsbehandling og hornhinneregenerering, inkludert limbal stamcelletransplantasjon, eksosomtransplantasjon og mitokondrietransplantasjon, er basert på studier som undersøkte hornhinnerespons på ulike skader samt behandlinger.
Derfor er det viktig å ha en standard, nøyaktig og pålitelig skademodell blant forskere. Flere teknikker og dyremodeller har blitt brukt til å indusere ulike hornhinneskader og undersøke ulike aspekter av hornhinnen sårhelingsprosessen. Alkalibrenningsmodellen er veletablert skademodell, som utføres ved å påføre natriumhydroksid direkte på hornhinnens overflate eller ved å bruke flatt filterpapir.
Vi utvikler en ny punch-trephine metode for å lage en musemodell av limbal stamcellemangel nøyaktig og pålitelig, overvinne begrensningene i tidligere teknikker, og minimere risikoen for utilstrekkelig skade på limbal området, eller ytterligere skade på fornix og conjunctiva. Denne teknikken har flere fordeler. Effektiv kjemisk skade på hele hornhinnens overflate og limbus hos mus, lokalisert og godt omskrevet skade på hornhinnen, påføring av væske i en forhåndsbestemt varighet, induksjon av forskjellige hornhinneskadestørrelser med egnede biopsistanser.
Det er også mulig for rotte- og kaninskademodeller.
