Em nossa equipe, estudamos os mecanismos celulares e moleculares que governam o desenvolvimento e reparo muscular. Na verdade, a distribuição especial de moléculas de mRNA é conhecida por regular vários processos celulares. E aqui, usamos a Drosophila como modelo para investigar como os mRNAs são especialmente distribuídos dentro do tecido muscular da mosca.
A dinâmica da expressão gênica tem sido comprovada para regular vários processos biológicos musculares. O advento de técnicas de sequenciamento de RNA de célula única e núcleo único de alto rendimento permitiu uma exploração abrangente da dinâmica transcricional. Uma limitação notável das técnicas ômicas clássicas é a incapacidade de fornecer a distribuição espacial das moléculas de RNAm dentro das fibras musculares de Drosophila.
Esta característica pode ser investigada por hibridização in situ por fluorescência de molécula única. Os métodos atuais são insuficientes para determinar a dinâmica transcricional do RNAm e a distribuição espacial dentro do sistema muscular Drosophila. Para resolver essa limitação, otimizamos um método para detectar e codificar as moléculas individuais de RNAm em fibras musculares de Drosophila com alta resolução espacial e escalas de moléculas sinalizadoras.
Um desafio atual no campo é visualizar a dinâmica de regeneração muscular em tempo real e em animais vivos. É por isso que estamos tentando desenvolver uma abordagem de imagem ao vivo para visualizar o comportamento das células-tronco musculares em seu ambiente nativo, e também para abordar a questão de seu modo e faixa de migração, por exemplo, durante a regeneração muscular e também sua diferenciação e isso com resolução sem precedentes.
