I vårt team studerade vi de cellulära och molekylära mekanismer som styr muskelutveckling och reparation. Faktum är att den speciella fördelningen av mRNA-molekyler är känd för att reglera olika cellulära processer. Och här använder vi Drosophila som modell för att undersöka hur mRNA är särskilt fördelade i flugans muskelvävnad.
Dynamik i genuttryck har visat sig reglera olika muskelbiologiska processer. Tillkomsten av RNA-sekvenseringstekniker med hög genomströmning av enskilda celler och enskilda kärnor har möjliggjort en omfattande utforskning av transkriptionsdynamiken. En anmärkningsvärd begränsning med klassiska omics-tekniker är en oförmåga att tillhandahålla den rumsliga fördelningen av mRNA-molekyler inom Drosophila-muskelfibrerna.
Denna egenskap kan undersökas genom fluorescens in situ-hybridisering med en enda molekyl. Nuvarande metoder är otillräckliga för att bestämma mRNA-transkriptionsdynamiken och den rumsliga fördelningen inom Drosophilas muskelsystem. För att ta itu med denna begränsning optimerade vi en metod för att detektera och kodifiera de enskilda mRNA-molekylerna i Drosophila-muskelfibrer med hög rumslig upplösning och signalmolekylskalor.
En aktuell utmaning inom området är att visualisera muskelregenereringsdynamiken i realtid och i levande djur. Det är därför vi försöker utveckla en metod för live-avbildning för att visualisera muskelstamcellernas beteende i deras naturliga miljö, och även för att ta itu med frågan om deras sätt och migrationsområde, till exempel under muskelregenereringen och även deras differentiering och detta med oöverträffad upplösning.
