Anvendelsen af de beskrevne metoder giver os mulighed for at holde et stort antal T. serrulatus-individer og giver os konsekvent forudsigelighed for antallet af skorpioner, der er nødvendige til årlig giftproduktion. Skorpioner er leddyr, der tilhører klassen Arachnida, orden Scorpiones, som omfatter ca. 2.621 arter 1,2. Disse dyr har et bredt geografisk område og er til stede på alle kontinenter undtagen Antarktis1. Forgiftning forårsaget af skorpioner resulterer i sygelighed eller død for tusindvis af mennesker hvert år globalt3. I 2019 blev det anslået, at der var mere end 1,2 millioner ulykker og 3,500 årlige dødsfald forårsaget af disse dyr. I Brasilien er antallet af sager steget eksponentielt og nået mere end 100,000 sager om året siden 2017 4,5,6. Den ukontrollerede urbanisering, der er observeret i Brasilien i de sidste årtier uden tilstrækkelig spildevandsbehandling og regelmæssig indsamling og bortskaffelse af affald, forbundet med miljøforringelse og klimaændringer, har givet betingelser for spredning af invasive skorpioner, såsom T. serrulatus, hvilket øger kontakten med mennesker og dermed resulterer i skadelige ulykker 4,7,8. Der er omkring 178 skorpionarter i Brasilien, men stik af medicinsk betydning er forårsaget af slægten Tityus, hvor fire arter (T. serrulatus, T. bahiensis, T. stigmurus og T. obscurus) er af medicinsk bekymring, hvor Tityus serrulatus er ansvarlig for de mest alvorlige tilfælde og dødsfald 7,9.
Instituto Butantan er det vigtigste laboratorium til fremstilling af modgift i Brasilien og leverer ca. 70.000 hætteglas med skorpionmodgift til det brasilianske sundhedssystem. Kort fortalt omfatter de trin, der er involveret i fremstilling af modgift, podning af giftafledte antigener i heste, indsamling og oprensning af rige antistofplasma, hvilket resulterer i 5 ml hætteglas med skorpionmodgift. Hvert hætteglas er i stand til at neutralisere mindst 1 mg skorpiongift pr. ml modgift. Arthropods Bioterium er en integreret del af industricentret, der er ansvarlig for at levere kildematerialet til skorpionmodgiften.
Arthropods Bioterium på Instituto Butantan blev oprindeligt navngivet Laboratory of Arthropods og blev etableret i 1967. Det var dog først i 1995, at laboratoriet flyttede til et eksklusivt anlæg dedikeret til at huse giftige leddyr10,11. På det tidspunkt blev skorpionerne holdt i et rum på 12 m2 uden kontrolleret temperatur og fordelt på ca. 13 indhegninger, der hver husede maksimalt 300 dyr10. På grund af den stigende efterspørgsel efter gift gennem årene og det deraf følgende behov for at øge antallet af dyr sammen med forbedringer i giftudvindingsprocessen skete der en større ændring i 2016. Laboratoriet blev integreret i det industrielle centrum af Instituto Butantan og blev omdøbt til Arthropods Bioterium. Derudover flyttede Bioterium i 2016 til et nyt specialbygget anlæg. Det nye skorpionrum dækker et areal på 24 m2 og indeholder ca. 48 polypropylenanlæg, der hver huser op til 300 dyr, som vist i figur 1. Dette resulterer i, at i alt 10.000 til 20.000 individer opretholdes under kontrollerede fangenskabsforhold med variationer i løbet af året. Skorpionerne forvaltes omhyggeligt under strenge opdrætsprotokoller for at sikre deres velfærd, samtidig med at de opfylder de høje produktionskrav og overholder god praksis og etiske standarder for dyrepleje. Til giftudvinding blev der designet og konstrueret et specifikt rum, der huser to luftstrømsskabe som vist i figur 2A. Disse skabe tjener som sikkerhedsforanstaltninger for at forhindre teknikere i at indånde giftpartikler under ekstraktionsproceduren. Personligt beskyttelsesudstyr (PPE) er obligatorisk for teknikerne, herunder syntetiske forklæder og FFP2-masker, for at undgå forurening og sikre deres sikkerhed.

Omkring 90% af T. serrulatus-skorpionerne i leddyrbioteriet stammer fra forskellige steder, indsamlet af kommunale sundhedsagenter som led i et overvågnings- og kontrolprogram for disse dyr. Skorpionerne fanges i byområder og sendes derefter til Instituto Butantan som en del af et samarbejdsprogram. T. serrulatus skorpioner er partenogenetiske, hvilket betyder, at fødslen opstår som følge af udviklingen af afkom fra ufrugtede æg uden tilstedeværelse af et individ af det modsatte køn12. På grund af denne type reproduktion fødes nogle dyr i fangenskab i vores anlæg og opretholdes, indtil de når voksenfasen, efter en lignende opdrætsprotokol som beskrevet nedenfor. Når de når voksenstadiet, føjes de til giftekstraktionsrutinen. Ved ankomsten gennemgår skorpionerne indledende screening, og de under gode sundhedsforhold opbevares i kollektive kabinetter, der indeholder højst 350 individer, divideret med deres oprindelse. For hvert kabinet udfyldes dagligt en kontrolformular, der giver data om fodringsdatoen og den type bytte, der tilbydes (cricket eller kakerlak), vandforsyning, rengøringsprocedurer, antallet af døde dyr og antallet af resterende levende inde i kabinettet.
I en forudbestemt dagsorden udsættes dyrene for elektrisk stimulering, og den resulterende gift fra hundredvis af individer frysetørres derefter, hvilket resulterer i partier frysetørret gift. Det standardiserede gift opnået ved denne proces er bestemt til hesteimmunisering som led i fremstillingsprocessen samt at tilvejebringe referencemateriale til kvalitetskontrol af det aktive stof og slutproduktet.
Der er meget få faciliteter i verden, der er i stand til at holde et så stort antal skorpioner og udføre mængden af giftekstraktioner i overensstemmelse med god fremstillingspraksis og etisk brug af dyrene, som det gøres i leddyrs bioterium fra Instituto Butantan. Derfor er vores mål at beskrive skorpionvedligeholdelsesprotokollerne og giftekstraktionsprocedurerne, der anvendes i T. serrulatus-husdyrhold , som med succes leverer den nødvendige mængde gift til produktion af skorpionmodgift.

captive maintenance and venom extraction of tityus serrulatus (brazilian yellow scorpion) for antivenom production

Skorpiongift er et folkesundhedsproblem i flere tropiske og subtropiske lande. Tityus serrulatus Lutz og Mello, 1922 (brasiliansk gul skorpion) er ansvarlige for ca. 150.000 gifttilfælde om året i Brasilien, hvoraf 10% kræver modgiftbehandling for at vende livstruende gifteffekter. Derfor opretholdes tusindvis af T. serrulatus-individer under kontrollerede fangenskabsbetingelser til giftekstraktion, der efterfølgende anvendes til fremstilling af den nationale forsyning af skorpionmodgift. Instituto Butantan er det vigtigste laboratorium til fremstilling af modgift i Brasilien og leverer omkring 70.000 hætteglas med skorpionmodgift til det brasilianske sundhedssystem. Således er opdrætsprotokollerne og giftekstraktionsmetoderne nøglepunkter for succesen med storstilet, standardiseret giftproduktion. Formålet med denne artikel er at beskrive protokollerne for opdræt af T. serrulatus , der omfatter opdrætsrutinen og giftudvindingsprocedurerne, følger god fremstillingspraksis og sikrer dyrevelfærd. Denne praksis giver mulighed for at opretholde op til 20.000 dyr i fangenskab med en rutine på 3.000 til 5.000 skorpioner, der malkes månedligt i henhold til efterspørgslen efter modgiftfremstilling, hvilket opnår et gennemsnit på 90% af positiv ekstraktion.

Scorpions are arthropods belonging to the class Arachnida, order Scorpiones, which comprises approximately 2,621 species1,2. These animals have a wide geographical range and are present on all continents except Antarctica1. Envenomation caused by scorpions results in the morbidity or death of thousands of people every year globally3. In 2019, it was estimated that there were more than 1.2 million accidents and 3,500 annual deaths caused by these animals. In Brazil, the number of cases has been increasing exponentially, reaching more than 100,000 cases per year since 20174,5,6. The uncontrolled urbanization observed in Brazil in the past decades, without adequate sewage treatment and regular collection and disposal of garbage, associated with environmental degradation and climate changes, has provided conditions for the proliferation of invasive scorpions, such as T. serrulatus, increasing the contact with humans and thus resulting in harmful accidents4,7,8. There are about 178 scorpion species in Brazil, but stings of medical importance are caused by the genus Tityus, with four species (T. serrulatus, T. bahiensis, T. stigmurus, and T. obscurus) being of medical concern, with Tityus serrulatus being responsible for the most severe cases and deaths7,9.
Instituto Butantan is the main antivenom-manufacturing laboratory in Brazil, providing approximately 70,000 vials of scorpion antivenom for the Brazilian health system. Briefly, the steps involved in antivenom manufacturing include the inoculation of venom-derived antigens in equines, the collection and purification of rich-antibodies plasma, resulting in 5 mL vials of scorpion antivenom. Each vial is capable of neutralizing, at least, 1 mg of scorpion venom per mL of antivenom. The Arthropods Bioterium is an integral part of the industrial center, responsible for supplying the source material for the scorpion antivenom.
The Arthropods Bioterium at Instituto Butantan was originally named the Laboratory of Arthropods and was established in 1967. However, it was only in 1995 that the Laboratory moved to an exclusive facility dedicated to housing venomous arthropods10,11. At that time, the scorpions were kept in a room of 12 m2 with no controlled temperature and distributed among approximately 13 enclosures, each housing a maximum of 300 animals10. Due to the increasing demand for venom over the years and the consequent need to increase the number of animals, along with improvements in the venom extraction process, a major change occurred in 2016. The Laboratory was integrated into the industrial center of Instituto Butantan and was renamed the Arthropods Bioterium. Additionally, in 2016, the Bioterium moved to a new purpose-built facility. The new scorpion room covers an area of 24 m2 and contains approximately 48 polypropylene enclosures, each housing up to 300 animals, as shown in Figure 1. This results in a total of 10,000 to 20,000 individuals being maintained under controlled captive conditions, with variations throughout the year. The scorpions are carefully managed under strict husbandry protocols to ensure their welfare while meeting the high production demands and adhering to good practices and ethical standards for animal care. For venom extraction, a specific room was designed and constructed, housing two airflow cabinets as shown in Figure 2A. These cabinets serve as safety measures to prevent technicians from inhaling venom particles during the extraction procedure. Personal protective equipment (PPE) is mandatory for the technicians, including synthetic aprons and FFP2 masks, to avoid any contamination and ensure their safety.

About 90% of the T. serrulatus scorpions in the Arthropods Bioterium originate from different sites, collected by municipal health agents as part of a surveillance and control program for these animals. The scorpions are caught in urban areas and then sent to Instituto Butantan as part of a collaborative program. T. serrulatus scorpions are parthenogenetic, meaning that birth occurs as a consequence of the development of offspring from unfertilized eggs, without the presence of an individual of the opposite sex12. Due to this type of reproduction, some animals are born in captivity in our facility and are maintained until they reach the adult phase, following a similar husbandry protocol as described below. Once they reach the adult stage, they are added to the venom extraction routine. Upon arrival, the scorpions undergo initial screening, and those in good health conditions are kept in collective enclosures containing a maximum of 350 individuals, divided by their origin. For each enclosure, a control form is filled out daily, providing data on the date of feeding and the type of prey offered (cricket or cockroach), water supply, cleaning procedures, the number of dead animals, and the number of remaining live ones inside the enclosure.
In a pre-scheduled agenda, animals are subjected to electric stimulation, and the resulting venom of hundreds of individuals is then lyophilized, resulting in batches of freeze-dried venom. The standardized venom obtained from this process is destined for horse immunization as part of the manufacturing process, as well as to provide reference material for quality control of the active substance and the final product.
There are very few facilities in the world capable of keeping such a large number of scorpions and performing the volume of venom extractions in accordance with good manufacturing practices and ethical use of the animals, as done in the Arthropods Bioterium of Instituto Butantan. Therefore, our objective is to describe the scorpion maintenance protocols and venom extraction procedures used in T. serrulatus husbandry, which successfully provide the necessary amount of venom for the production of scorpion antivenom.

This protocol describes the method to maintain scorpions for the extraction of high amounts of venom, which is used in the production of neutralizing equine-delivered immunoglobulins or antivenoms. There is a high demand for scorpion antivenom in tropical countries, which is the keystone for treating human envenomation cases. The maintainancy procedures reproduce optimal natural conditions, keeping the scorpions in captivity, thus with considering animal welfare and good laboratory practice.In parallel, venom extraction is based on electrical stimulation in the scorpion telson, with minimal harmful effects on the animals. This protocol enabled us to increase exponentially the number of animals maintained in captivity, ranging from 4, 000 to 20, 000 individuals in 10 years. At the same time, venom has been extracted sufficiently to immunize horses to obtain plasma with the specific antibodies, which is the starting material for the manufacturing process that results in around 100, 000 vials of anti-venom per year.The venom milking procedures different from others described in the literature in which the telson is removed, thus limiting the number of extraction per animal. The accumulated experience of the technicians of our facility allow us to extract venom from 200 to 300 animals in around 1 to 1 1/2 hours without anesthetizing the animals or making other invasive procedures.

Tredive standard operationelle procedurer (SOP'er) blev udviklet for hver procedure, hvilket sikrer repeterbarheden af procedurerne blandt teknikere og overholdelse af kvalitetsparametre. Denne protokol beskriver metoden til at opretholde skorpioner til ekstraktion af store mængder gift, som anvendes til fremstilling af neutraliserende hesteleverede immunglobuliner eller modgifte. Der er stor efterspørgsel efter skorpionmodgift i tropiske lande, som er slutstenen til behandling af humane gifttilfælde. Vedligeholdelsesprocedurerne reproducerer optimale naturlige forhold og holder skorpionerne i fangenskab, således med hensyntagen til dyrevelfærd og god laboratoriepraksis.Parallelt er giftekstraktion baseret på elektrisk stimulering i skorpionens telson, med minimale skadelige virkninger på dyrene. Denne protokol gjorde det muligt for os eksponentielt at øge antallet af dyr, der holdes i fangenskab, fra 4.000 til 20.000 individer på 10 år. Samtidig er gift blevet ekstraheret tilstrækkeligt til at immunisere heste for at opnå plasma med de specifikke antistoffer, som er udgangsmaterialet til fremstillingsprocessen, der resulterer i omkring 100.000 hætteglas med antigift om året.Giftmalkeprocedurerne adskiller sig fra andre beskrevet i litteraturen, hvor telsonen fjernes, hvilket begrænser antallet af ekstraktioner pr.
