Avantages et inconvénients de divers paradigmes de lésions rétiniennes chez les têtards de Xénope

Des millions de personnes dans le monde sont atteintes de diverses maladies dégénératives de la rétine conduisant à la cécité, telles que la rétinite pigmentaire, la rétinopathie diabétique ou la dégénérescence maculaire liée à l’âge (DMLA). À ce jour, ces affections restent en grande partie incurables. Les approches thérapeutiques actuellement à l’étude comprennent la thérapie génique, les transplantations de cellules ou de tissus, les traitements neuroprotecteurs, l’optogénétique et les prothèses. Une autre stratégie émergente est basée sur l’auto-régénération par l’activation de cellules endogènes ayant un potentiel de cellules souches. Les cellules gliales de Müller, le principal type de cellules gliales de la rétine, font partie des sources cellulaires d’intérêt dans ce contexte. En cas de blessure, ils peuvent se dédifférencier, proliférer et générer des neurones 1,2,3. Bien que ce processus soit très efficace chez le poisson-zèbre ou le Xénope, il est largement inefficace chez les mammifères.
Néanmoins, il a été démontré que des traitements appropriés avec des protéines mitogènes ou une surexpression de divers facteurs peuvent induire la réentrée dans le cycle des cellules gliales de Müller chez les mammifères et, dans certains cas, déclencher leur engagement neurogénique ultérieur 1,2,3,4,5. Cela reste cependant largement insuffisant pour les traitements. Par conséquent, il est nécessaire d’accroître nos connaissances sur les mécanismes moléculaires sous-jacents à la régénération afin d’identifier des molécules capables de transformer efficacement les propriétés des cellules souches de Müller en nouvelles stratégies thérapeutiques cellulaires.
Dans ce but, nous avons développé plusieurs paradigmes de lésions dans Xenopus qui déclenchent la dégénérescence des cellules rétiniennes. Ici, nous présentons (1) une lésion mécanique de la rétine qui n’est pas spécifique à un type de cellule, (2) un modèle d’ablation cellulaire conditionnelle et réversible utilisant le système NTR-MTZ qui cible les cellules en bâtonnets, (3) un knock-out de la rhodopsine médié par CRISPR/Cas9, un modèle de rétinite pigmentaire qui déclenche une dégénérescence progressive des cellules en bâtonnet, et (4) un CoCl2-modèle cytotoxique induit qui, selon la dose, peut cibler spécifiquement les cônes ou conduire à une dégénérescence plus large des cellules rétiniennes. Nous mettons en évidence les particularités, les avantages et les inconvénients de chaque paradigme.

Les maladies neurodégénératives de la rétine sont les principales causes de cécité. Parmi les nombreuses stratégies thérapeutiques explorées, la stimulation de l’autoréparation s’est récemment révélée particulièrement attrayante. Une source cellulaire d’intérêt pour la réparation de la rétine est la cellule gliale de Müller, qui abrite un potentiel de cellules souches et une capacité de régénération extraordinaire chez les anamniotes. Ce potentiel est cependant très limité chez les mammifères. L’étude des mécanismes moléculaires sous-jacents à la régénération rétinienne dans des modèles animaux dotés de capacités de régénération devrait permettre de mieux comprendre comment libérer la capacité latente des cellules de Müller chez les mammifères à régénérer la rétine. Il s’agit d’une étape clé pour le développement de stratégies thérapeutiques en médecine régénérative. Dans ce but, nous avons développé plusieurs paradigmes de lésions rétiniennes dans Xenopus : une lésion mécanique de la rétine, une lignée transgénique permettant l’ablation conditionnelle des photorécepteurs médiée par la nitroréductase, un modèle de rétinite pigmentaire basé sur le knock-out de la rhodopsine médié par CRISPR/Cas9, et un modèle cytotoxique piloté par des injections intraoculaires de CoCl2 . En mettant en évidence leurs avantages et leurs inconvénients, nous décrivons ici cette série de protocoles qui génèrent diverses conditions dégénératives et permettent l’étude de la régénération rétinienne chez Xenopus.

Millions of people worldwide are afflicted with various retinal degenerative diseases leading to blindness, such as retinitis pigmentosa, diabetic retinopathy, or age-related macular degeneration (AMD). To date, these conditions remain largely untreatable. Current therapeutic approaches under evaluation include gene therapy, cell or tissue transplantations, neuroprotective treatments, optogenetics, and prosthetic devices. Another emerging strategy is based on self-regeneration through the activation of endogenous cells with stem cell potential. M&#252;ller glial cells, the major glial cell type of the retina, are among cellular sources of interest in this context. Upon injury, they can dedifferentiate, proliferate, and generate neurons1,2,3. Although this process is very effective in zebrafish or Xenopus, it is largely inefficient in mammals.
Nonetheless, it has been shown that appropriate treatments with mitogenic proteins or overexpression of various factors can induce mammalian M&#252;ller glia cell-cycle re-entry and, in some cases, trigger their subsequent&#160;neurogenesis commitment1,2,3,4,5. This remains, however, largely insufficient for treatments. Hence, increasing our knowledge of the molecular mechanisms underlying regeneration is necessary to identify molecules able to efficiently turn M&#252;ller stem-like cell properties into new cellular therapeutic strategies.
With this aim, we developed several injury paradigms in Xenopus that trigger retinal cell degeneration. Here, we present (1) a mechanical retinal injury that is not cell type-specific, (2) a conditional and reversible cell ablation model using the NTR-MTZ system that targets rod cells, (3) a CRISPR/Cas9-mediated rhodopsin knockout, a model of retinitis pigmentosa that triggers progressive rod cell degeneration, and (4) a CoCl2-induced cytotoxic model that according to the dose can specifically target cones or lead to broader retinal cell degeneration. We highlight the particularities, advantages, and disadvantages of each paradigm.

My lab works on retinal regeneration. We are using the Xenopus frog as a model system. This amphibian is indeed quite fascinating because unlike mammals, like us, it can regenerate its retina very efficiently in case of injury.And we are studying the underlying mechanism because, in the future, it could be useful to trigger retinal regeneration in human patients afflicted with retinal neurodegenerative diseases. We have recently discovered that in addition to stem cells in the periphery of the retina and the retinal pigmented epithelium, neuroglial cells can also be recruited for retinal regeneration in case of injury. And so we are now studying the links between neuroinflammation and the regenerative capacity of these cells.Indeed, it seems that the neuroinflammatory niche is a key player in the modulation of the regeneration of the retina. To study the cellular and molecular mechanisms involved in retinal regeneration, we developed several retinal injury paradigms in Xenopus. The first is a mechanical retinal injury.The second is a transgenic line allowing for nitroreductase-mediated photoreceptor conditional ablation. The third is a retinitis pigmentosa model based on CRISPR/Cas9-mediated rhodopsin knockout, and, to finish, a cytotoxic model driven by intraocular injection of cobalt chloride or CoCl2. My host laboratory has showed that although Xenopus can regenerate its retina, the efficiency is highly variable and depends on the stages of the tadpole or on species, Xenopus laevis or tropicalis.This makes Xenopus a fantastic model for illustrating the molecular mechanisms that trigger or limit retinal regeneration.

Mon laboratoire travaille sur la régénération rétinienne. Nous utilisons la grenouille Xénope comme système modèle. Cet amphibien est en effet assez fascinant car contrairement aux mammifères, comme nous, il peut régénérer sa rétine très efficacement en cas de blessure.Et nous étudions le mécanisme sous-jacent car, à l’avenir, il pourrait être utile de déclencher la régénération rétinienne chez les patients humains atteints de maladies neurodégénératives rétiniennes. Nous avons récemment découvert qu’en plus des cellules souches à la périphérie de la rétine et de l’épithélium pigmenté de la rétine, les cellules neurogliales peuvent également être recrutées pour la régénération rétinienne en cas de lésion. Nous étudions donc maintenant les liens entre la neuroinflammation et la capacité de régénération de ces cellules.En effet, il semblerait que la niche neuro-inflammatoire soit un acteur clé dans la modulation de la régénération de la rétine. Pour étudier les mécanismes cellulaires et moléculaires impliqués dans la régénération rétinienne, nous avons développé plusieurs paradigmes de lésions rétiniennes chez Xenopus.
