Kenmerkend angstgedrag dat bij NTT is waargenomen, is positief gecorreleerd met serotoninespiegels die in de hersenen zijn geanalyseerd21. Bijvoorbeeld, na blootstelling aan para-chloorfenylalanine (PCPA), een remmer van 5-HT biosynthese, vertoonden vissen positieve geotaxis en verlaagde 5-HT-niveaus in de hersenen22, resultaten die sterk lijken op die verkregen met METH. Daarom suggereert de afname van de serotoninespiegels in de hersenen en de weergave van positieve geota...

De zebravis (Danio rerio) is een populaire modelsoort van gewervelde dieren voor ecotoxicologie, ontdekking van geneesmiddelen en veiligheidsfarmacologische studies. De goedkope, gevestigde moleculair genetische hulpmiddelen en het behoud van belangrijke fysiologische processen die betrokken zijn bij de morfogenese en het onderhoud van het zenuwstelsel maken zebravissen tot een ideaal diermodel voor neurowetenschappelijk onderzoek, inclusief neurogedragstoxicologie 1,2. Het belangrijkste eindpunt voor het evalueren van de neurotoxiciteit van een chemische stof was tot voor kort de aanwezigheid van neuropathologische effecten. De laatste tijd zijn gedragseindpunten echter opgenomen in neurotoxicologische screeningprotocollen, en deze functionele uitkomsten worden nu vaak gebruikt om de potentiële neurotoxiciteit van chemicaliën te identificeren en te bepalen. Bovendien zijn gedragseindpunten zeer relevant vanuit ecologisch oogpunt, aangezien zelfs een zeer milde gedragsverandering bij vissen het voortbestaan van het dier in natuurlijke omstandigheden in gevaar kan brengen.
Een van de meest gebruikte gedragstesten in het onderzoek naar volwassen zebravissen is de nieuwe tanktest (NTT), die angstachtig gedrag meet 6,7. In deze test worden vissen blootgesteld aan nieuwigheid (vissen worden in een onbekend aquarium geplaatst), een milde aversieve stimulus en hun gedragsreacties worden geobserveerd. NTT wordt voornamelijk gebruikt om basale locomotorische activiteit, geotaxis, bevriezing en grillige bewegingen van vissen te beoordelen. Grillig8 wordt gekenmerkt door abrupte richtingsveranderingen (zigzaggen) en herhaalde episodes van versnellingen (darten). Het is een alarmreactie en wordt meestal waargenomen voor of na bevriezingsepisodes. Bevriezingsgedrag komt overeen met een volledige stopzetting van de bewegingen van de vis (behalve operculaire en oculaire bewegingen) op de bodem van de tank, in tegenstelling tot immobiliteit veroorzaakt door sedatie, die hypolocobeweging, akinesie en zinken veroorzaakt8. Bevriezing is meestal gerelateerd aan een hoge staat van stress en angst en maakt ook deel uit van onderdanig gedrag. Complex gedrag is een uitstekende indicator van de staat van angst van dieren. Van NTT is aangetoond dat het gevoelig is voor farmacologische en genetische manipulatie9, waardoor het een waardevol hulpmiddel is voor het bestuderen van de neurale basis van angst en aanverwante aandoeningen.
Zebravissen zijn een zeer sociale soort, dus we kunnen een breed scala aan sociaal gedrag meten. De ondieptetest (ST) en de sociale voorkeurstest (SPT) zijn de meest gebruikte tests om sociaal gedrag te beoordelen10. De ST meet de neiging van vissen om11 te groeperen door hun ruimtelijk gedrag en bewegingspatronen te kwantificeren. ST is nuttig voor het bestuderen van groepsdynamiek, leiderschap, sociaal leren en het begrijpen van het sociale gedrag van veel vissoorten12. De SPT in volwassen zebravissen werd aangepast van Crawley's voorkeur voor sociale nieuwheidstest voor muizen13 en werd al snel een populaire gedragstest voor de studie van sociale interactie in deze modelsoort14. Deze twee tests zijn ook aangepast voor gebruik in drugsscreeningstests en zijn veelbelovend gebleken voor het identificeren van nieuwe verbindingen die sociaal gedrag moduleren15,16.
Over het algemeen zijn gedragstesten bij volwassen zebravissen krachtige hulpmiddelen die waardevolle informatie kunnen opleveren over de gedragsmechanismen of de neurofenotypes van actieve stoffen en misbruikte medicijnen. Dit protocol beschrijft hoe deze gedragshulpmiddelen7 kunnen worden geïmplementeerd met elementaire materiële middelen en hoe ze kunnen worden toegepast in toxiciteitstests om de effecten van een breed scala aan neuroactieve stoffen te karakteriseren. Daarnaast zullen we zien dat dezelfde tests kunnen worden toegepast om de neurologische gedragseffecten van acute blootstelling aan een neuroactieve stof (methamfetamine) te beoordelen, maar ook om deze effecten te karakteriseren na chronische blootstelling aan omgevingsconcentraties van een pesticide (glyfosaat).

neurotoxicity assessment in adult danio rerio using a battery of behavioral tests in a single tank

De aanwezigheid van neuropathologische effecten bleek jarenlang het belangrijkste eindpunt te zijn voor de beoordeling van de neurotoxiciteit van een chemische stof. In de afgelopen 50 jaar zijn de effecten van chemicaliën op het gedrag van modelsoorten echter actief onderzocht. Geleidelijk aan werden gedragseindpunten opgenomen in neurotoxicologische screeningsprotocollen, en deze functionele uitkomsten worden nu routinematig gebruikt om de potentiële neurotoxiciteit van chemicaliën te identificeren en te bepalen. Gedragstesten bij volwassen zebravissen bieden een gestandaardiseerde en betrouwbare manier om een breed scala aan gedragingen te bestuderen, waaronder angst, sociale interactie, leren, geheugen en verslaving. Gedragstesten bij volwassen zebravissen omvatten meestal het plaatsen van de vissen in een experimentele arena en het registreren en analyseren van hun gedrag met behulp van videotrackingsoftware. Vissen kunnen worden blootgesteld aan verschillende stimuli en hun gedrag kan worden gekwantificeerd met behulp van verschillende statistieken. De nieuwe tanktest is een van de meest geaccepteerde en meest gebruikte tests om angstachtig gedrag bij vissen te bestuderen. De scholen- en sociale voorkeurstests zijn nuttig bij het bestuderen van het sociale gedrag van zebravissen. Deze test is vooral interessant omdat het gedrag van de hele school wordt bestudeerd. Deze tests hebben bewezen zeer reproduceerbaar en gevoelig te zijn voor farmacologische en genetische manipulaties, waardoor ze waardevolle hulpmiddelen zijn voor het bestuderen van de neurale circuits en moleculaire mechanismen die ten grondslag liggen aan gedrag. Bovendien kunnen deze tests worden gebruikt bij het screenen van geneesmiddelen om verbindingen te identificeren die potentiële gedragsmodulatoren kunnen zijn.
We zullen in dit werk laten zien hoe gedragsinstrumenten kunnen worden toegepast in de neurotoxicologie van vissen, waarbij we het effect van methamfetamine, een recreatieve drug, en glyfosaat, een milieuverontreinigende stof, analyseren. De resultaten tonen de significante bijdrage aan van gedragstesten in volwassen zebravissen aan het begrip van de neurotoxicologische effecten van milieuverontreinigende stoffen en medicijnen, naast het verschaffen van inzicht in de moleculaire mechanismen die de neuronale functie kunnen veranderen.

The zebrafish (Danio rerio) is a popular model vertebrate species for ecotoxicology, drug discovery, and safety pharmacology studies. Its low cost, well-established molecular genetic tools, and conservation of key physiological processes involved in the morphogenesis and maintenance of the nervous system make zebrafish an ideal animal model for neuroscience research, including neurobehavioral toxicology1,2. The main endpoint for evaluating the neurotoxicity of a chemical was, until recently, the presence of neuropathological effects. Lately, however, behavioral endpoints have been incorporated into neurotoxicological screening protocols, and these functional outcomes are now commonly used to identify and determine the potential neurotoxicity of chemicals3,4. Moreover, behavioral endpoints are highly relevant from an ecological point of view, as even a very mild behavioral change in fish could endanger the survival of the animal in natural conditions5.
One of the most used behavioral assays in adult zebrafish research is the novel tank test (NTT), which measures anxiety-like behavior6,7. In this assay, fish are exposed to novelty (fish are placed in an unfamiliar tank), a mild aversive stimulus and their behavioral responses are observed. NTT is used to assess basal locomotor activity, geotaxis, freezing, and erratic movements of fish, principally. Erratic8 is characterized by abrupt changes of direction (zigzagging) and repeated episodes of accelerations (darting). It is an alarm reaction and is usually observed before or after freezing episodes. Freezing behavior corresponds to a complete cessation of the fish&#39;s movements (except for opercular and ocular movements) while on the bottom of the tank, as distinguished from immobility caused by sedation, which causes hypolocomotion, akinesia, and sinking8. Freezing is usually related to a high state of stress and anxiety and is also part of submissive behavior. Complex behaviors are excellent indicators of the state of anxiety of animals. NTT has been shown to be sensitive to pharmacological and genetic manipulation9, making it a valuable tool for studying the neural basis of anxiety and related disorders.
Zebrafish are a highly social species, so we can measure a wide range of social behaviors. The shoaling test (ST) and the social preference test (SPT) are the most used assays to assess social behavior10. The ST measures the tendency of fish to group together11 by quantifying their spatial behavior and movement patterns. ST is useful for studying group dynamics, leadership, social learning, and understanding the social behavior of many fish species12. The SPT in adult zebrafish was adapted from Crawley&#39;s preference for social novelty test for mice13&#160;and quickly became a popular behavioral assay for the study of social interaction in this model species14. These two tests have also been adapted for use in drug screening assays and have shown promise for identifying novel compounds that modulate social behavior15,16.
In general, behavioral assays in adult zebrafish are powerful tools that can provide valuable information on the behavior mechanisms or the neurophenotypes of active compounds and abused drugs17. This protocol details how to implement these behavioral tools7 with basic material resources and how to apply them in toxicity assays to characterize the effects of a wide range of neuroactive compounds. In addition, we will see that the same tests can be applied to assess the neurobehavioral effects of acute exposure to a neuroactive compound (methamphetamine) but also to characterize these effects after chronic exposure to environmental concentrations of a pesticide (glyphosate).

Author Spotlight: Assessment of Environmental and Drug-Induced Behavioral Changes in Adult Zebrafish

Our main research aims to identify the behavioral effects induced by environmental pollutants and drugs in fish. Also trying to understand the mechanisms leading to these effects. A social and anxiety-like behavior cannot be conveniently addressed using embryos.We conduct studies targeting these behaviors in the adult Zebrafish model. This protocol details how to implement behavioral tools with basic material resources to facilitate the reproducibility of tests and harmonize the results obtained within the scientific community. The assays presented in this protocol have proven to be highly reproducible and sensitive to genetic and pharmacological manipulations, making them variable and noninvasive tools fostering the neural and molecular mechanisms underlying behavior.Once a behavioral effort has been identified, the most challenging part is to determine the specific modes of action leading to the observative efforts. Starting by the molecular initiating event where predictions can provide some clues on the molecular initiating event, we are still far from obtaining accurate predictions. We are currently interested in the determining the phase validity of the chemical models of Parkinson's disease developed in embryonic and adult Zebrafish by using MPTP and rotenone with the same, the kinematic of the gait-like movement or during an explosive movement like the acoustic evoked escape response, will be evaluated.

In deze sectie zullen we kijken naar enkele mogelijke toepassingen van deze gedragshulpmiddelen in de neurotoxicologie van vissen. Ons belangrijkste onderzoek is gericht op het identificeren van de gedragseffecten die worden veroorzaakt door milieuverontreinigende stoffen en medicijnen bij vissen. Ook proberen we de mechanismen te begrijpen die tot deze effecten leiden. Een sociaal en angstachtig gedrag kan niet gemakkelijk worden aangepakt met behulp van embryo's.We voeren studies uit die gericht zijn op dit gedrag in het volwassen zebravismodel. Dit protocol beschrijft hoe gedragsinstrumenten kunnen worden geïmplementeerd met elementaire materiële middelen om de reproduceerbaarheid van tests te vergemakkelijken en de verkregen resultaten binnen de wetenschappelijke gemeenschap te harmoniseren. De tests die in dit protocol worden gepresenteerd, hebben bewezen zeer reproduceerbaar en gevoelig te zijn voor genetische en farmacologische manipulaties, waardoor ze variabele en niet-invasieve hulpmiddelen zijn die de neurale en moleculaire mechanismen die ten grondslag liggen aan gedrag bevorderen.Zodra een gedragsinspanning is geïdentificeerd, is het meest uitdagende deel het bepalen van de specifieke werkingsmechanismen die leiden tot de observerende inspanningen. Beginnend met de moleculaire initiërende gebeurtenis, waar voorspellingen enkele aanwijzingen kunnen geven over de moleculaire initiërende gebeurtenis, zijn we nog ver verwijderd van het verkrijgen van nauwkeurige voorspellingen. We zijn momenteel geïnteresseerd in het bepalen van de fasevaliditeit van de chemische modellen van de ziekte van Parkinson ontwikkeld in embryonale en volwassen zebravissen door gebruik te maken van MPTP en rotenon met dezelfde, de kinematica van de loopachtige beweging of tijdens een explosieve beweging zoals de akoestisch opgewekte ontsnappingsreactie, zal worden geëvalueerd.
