DNA-skaderespons er uundværlig for livet. Da DNA konstant udsættes for forskellige skadelige motorer, fører manglende reparation af dem til genomers ustabilitet og sygdomme. Vi udviklede en metode baseret på RPA2's biologiske egenskaber til at visualisere enkeltstrenget DNA-kanal under DNA-reparation i forskellige cellecyklusstadier.
Traditionel metode til enkeltstrenget DNA-detektion bruger BrdU-antistoffer. Denne tilgang kan bruges til aktivt at replikere celler. Derudover har dette antistof tendens til at krydsreagere med andre nukleotidanaloger, hvilket begrænser deres anvendelse.
I modsætning til BRDU har vores tilgang et højere signal / støjforhold, hvilket gør det anvendeligt i hver cyklusfase. Forståelse af virkningen af forskellige DNA-reparationsveje i forskellige cellecyklusfaser og potentielt ikke-replikerende celler er afgørende. Vores protokol giver en ny tilgang og værktøj til at visualisere enkeltstrengede DNA-foci, hvor klassiske tilgange, der bruger BrdU, er begrænsede.
En omfattende forståelse af DNA-reparationsprocesser kan tjene som terapeutiske mål til behandling af kræft, aldring og neurologiske sygdomme. De fleste af vores celler er terminalt differentierede og replikerer og deler sig derfor ikke. Det er afgørende at få en bedre forståelse af, hvordan disse celler reparerer forskellige DNA-læsioner.
Derfor vil vores laboratorium primært fokusere på postreplikative DNA-reparationsprocesser og udvikle nye værktøjer til at studere dem.
