DNA-skaderespons er uunnværlig for livet. Siden DNA hele tiden blir utsatt for forskjellige skadelige motorer, fører mangel på reparasjon av dem til genomenes ustabilitet og sykdommer. Vi utviklet en metode basert på den biologiske egenskapen til RPA2, for å visualisere enkeltstrenget DNA-kanal under DNA-reparasjon i forskjellige cellesyklusstadier.
Tradisjonell metode for enkeltstrengs DNA-deteksjon bruker BrdU-antistoffer. Denne tilnærmingen kan brukes til aktivt replikerende celler. I tillegg har dette antistoffet en tendens til å kryssreagere med andre nukleotidanaloger, noe som begrenser bruken av dem.
I motsetning til BRDU har tilnærmingen vår et høyere signal/støy-forhold, noe som gjør den brukbar i hver syklusfase. Å forstå virkningen av forskjellige DNA-reparasjonsveier i forskjellige cellesyklusfaser og potensielt ikke-replikerende celler er avgjørende. Vår protokoll gir en ny tilnærming og et verktøy for å visualisere enkeltstrengede DNA-foci, der klassiske tilnærminger som bruker BrdU er begrenset.
En omfattende forståelse av DNA-reparasjonsprosesser kan tjene som terapeutiske mål for å behandle kreft, aldring og nevrologiske sykdommer. De fleste av cellene våre er terminalt differensiert og replikerer derfor ikke og deler seg. Det er avgjørende å få en bedre forståelse av hvordan disse cellene reparerer ulike DNA-lesjoner.
Derfor vil vårt laboratorium hovedsakelig fokusere på postreplikerende DNA-reparasjonsprosesser og utvikle nye verktøy for å studere dem.
