Реакция на повреждение ДНК необходима для жизни. Поскольку ДНК постоянно подвергается воздействию различных повреждающих двигателей, отсутствие их репарации приводит к нестабильности геномов и заболеваниям. Мы разработали метод, основанный на биологическом свойстве RPA2, для визуализации одноцепочечного тракта ДНК во время репарации ДНК на разных стадиях клеточного цикла.
Традиционный метод обнаружения одноцепочечной ДНК использует антитела BrdU. Этот подход может быть использован для активно реплицирующихся клеток. Кроме того, эти антитела имеют тенденцию к перекрестной реакции с другими аналогами нуклеотидов, что ограничивает их использование.
В отличие от BRDU, наш подход имеет более высокое соотношение сигнал/шум, что делает его пригодным для использования на каждой фазе цикла. Понимание действия различных путей репарации ДНК в различных фазах клеточного цикла и потенциально нереплицирующихся клеток имеет решающее значение. Наш протокол предоставляет новый подход и инструмент для визуализации одноцепочечных очагов ДНК, в то время как классические подходы, использующие BrdU, ограничены.
Всестороннее понимание процессов репарации ДНК может служить терапевтическими мишенями для лечения рака, старения и неврологических заболеваний. Большинство наших клеток терминально дифференцированы и поэтому не размножаются и не делятся. Очень важно лучше понять, как эти клетки восстанавливают различные повреждения ДНК.
Поэтому наша лаборатория будет в основном заниматься пострепликативными процессами репарации ДНК и разрабатывать новые инструменты для их изучения.
