El cristalino ocular es un tejido flexible y transparente situado en la región anterior del ojo que funciona para enfocar la luz en la retina. La capacidad de funcionamiento de la lente se puede atribuir, en parte, a su intrincada arquitectura y organización 1,2,3,4,5,6. Alrededor del tejido del cristalino se encuentra la cápsula, una membrana basal esencial para mantener la estructura del cristalino y las propiedades biomecánicas 7,8,9. El cristalino en sí es completamente celular, y consta de dos tipos de células: epiteliales y fibrosas. La capa epitelial está formada por una monocapa de células cuboidales que recubren el hemisferio anterior del cristalino10. A lo largo de la vida, las células epiteliales proliferan y migran a lo largo de la cápsula del cristalino hacia el ecuador del cristalino. Las células epiteliales anteriores están en reposo y en sección transversal, y cerca del ecuador del cristalino, las células epiteliales proliferan y comienzan a experimentar el proceso de diferenciación en nuevas células de fibra11,12. Las células epiteliales ecuatoriales se transforman de células empaquetadas al azar en filas meridionales organizadas con células en forma de hexágono. La forma de las células hexagonales se mantiene en el lado basal de estas células diferenciadoras, mientras que el lado apical se contrae y se ancla en el punto de apoyo del cristalino o modiolo 4,13,14,15. A medida que las células epiteliales ecuatoriales comienzan a alargarse en células fibrosas recién formadas, las puntas apicales de las células migran a lo largo de la superficie apical de las células epiteliales anteriores hacia el polo anterior, mientras que las puntas basales se mueven a lo largo de la cápsula del cristalino hacia el polo posterior. Las nuevas generaciones de células de fibra se superponen a las generaciones anteriores de células, creando capas esféricas de fibras. Durante el proceso de elongación y maduración celular, las células de fibra alteran sustancialmente su morfología 11,12,16. Estas células de fibra forman la mayor parte de la masa del cristalino 11,12,16,17,18.
Los mecanismos moleculares que contribuyen al establecimiento de las intrincadas microestructuras del cristalino, la morfología celular y la organización celular única no se conocen del todo. Además, la contribución de la cápsula del cristalino y la estructura celular a la función general del cristalino (transparencia, cambio de forma del cristal) no está clara. Sin embargo, estas relaciones están siendo dilucidadas utilizando una nueva metodología de imagen y evaluaciones cuantitativas de las características estructurales y celulares del cristalino 2,4,19,20,21,22. Se han desarrollado nuevos protocolos para obtener imágenes de lentes completas que permiten la visualización de alta resolución espacial de la cápsula del cristalino, las células epiteliales y las células de fibra periférica. Esto incluye la metodología para cuantificar el grosor de la cápsula, el tamaño de la célula, el tamaño y la circularidad del núcleo celular, el orden de las hileras meridionales, el empaquetamiento de las células de fibra y el ancho de las células de fibra. Estos métodos de visualización y cuantificación de imágenes permiten un examen morfométrico en profundidad y tienen ventajas sobre otros métodos de visualización (imágenes de montajes planos o secciones de tejido) al preservar la estructura general del tejido en 3D. Estos métodos han permitido probar nuevas hipótesis y permitirán un avance continuo en la comprensión del desarrollo y la función del patrón de las células del cristalino.
Para los siguientes experimentos, utilizamos ratones de tipo salvaje y Rosa26-tdTomato tándem dímero-Tomate (B6.129(Cg)-Gt(ROSA) (tdTomato)23 (Jackson Laboratories) en el fondo C57BL/6J entre las edades de 6 y 10 semanas, de ambos sexos. Los ratones tdTomato permiten la visualización de las membranas plasmáticas celulares en lentes vivas a través de la expresión de la proteína tdTomato fusionada con los aminoácidos N-terminal 8 de una proteína MARCKS mutada que se dirige a la membrana plasmática a través de la miritilación N-terminal y los sitios internos de cisteína-palmitoilación23. También utilizamos ratones NMIIAE1841K/E1841K 24 obtenidos originalmente del Dr. Robert Adelstein (Instituto Nacional del Corazón, los Pulmones y la Sangre, Institutos Nacionales de Salud, Bethesda, MD). Como se describió anteriormente20, los ratones NMIIAE1841K/E1841K en el fondo FvBN/129SvEv/C57Bl6 que tienen pérdida de la proteína de filamento intermedio con perlas CP49 (mantiene la morfología de la célula de fibra madura y la biomecánica del cristalino completo), se retrocruzan con ratones de tipo salvaje C57BL6/J. Examinamos a las crías para detectar la presencia del alelo CP49 de tipo salvaje. 
Las imágenes confocales se realizaron en un microscopio de fluorescencia confocal de barrido láser con un aumento de 20x (NA = 0,8, distancia de trabajo = 0,55 mm, zoom 1x), 40x (NA = 1,3 objetivo de aceite, distancia de trabajo = 0,2 mm, zoom de 1x) o 63x (NA = objetivo de aceite de 1,4, distancia de trabajo = 0,19 mm, zoom de 1x). Todas las imágenes se adquirieron utilizando un tamaño estenopeico, que es un determinante del espesor de la sección óptica, a 1 unidad de Airy (los espesores ópticos resultantes se indican en las leyendas de las figuras). Las imágenes fueron procesadas en Zen Software. Las imágenes se exportaron a .tif formato y luego se importaron al software FIJI ImageJ (imageJ.net).

El cristalino ocular es un tejido transparente y flexible que altera su forma para enfocar la luz desde diferentes distancias en la retina. The ocular lens is a flexible, transparent tissue situated at the anterior region of the eye that functions to fine-focus light onto the retina. The ability of the lens to function can be attributed, in part, to its intricate architecture and organization1,2,3,4,5,6. Surrounding the lens tissue is the capsule, a basement membrane essential for maintaining lens structure and biomechanical properties7,8,9. The lens itself is entirely cellular, consisting of two cell types: epithelial and fiber cells. The epithelial layer consists of a monolayer of cuboidal cells that cover the anterior hemisphere of the lens10. Throughout life, the epithelial cells proliferate and migrate along the lens capsule toward the lens equator. Anterior epithelial cells are quiescent and cobble-stone in cross-section, and near the lens equator, epithelial cells proliferate and start to undergo the differentiation process into new fiber cells11,12. Equatorial epithelial cells transform from randomly packed cells into organized meridional rows with hexagon-shaped cells. Hexagonal cell shape is maintained on the basal side of these differentiating cells while the apical side constricts and anchors at the lens fulcrum or modiolus4,13,14,15. As the equatorial epithelial cells start to elongate into newly formed fiber cells, the apical tips of the cells migrate along the apical surface of anterior epithelial cells toward the anterior pole while the basal tips move along the lens capsule toward the posterior pole. New generations of fiber cells overlay previous generations of cells, creating spherical shells of fibers. During the cell elongation and maturation process, fiber cells substantially alter their morphology11,12,16. These fiber cells form the bulk of the lens mass11,12,16,17,18.
The molecular mechanisms that contribute to establishing intricate lens microstructures, cell morphology, and unique cellular organization are not entirely known. Moreover, the contribution of the lens capsule and cell structure to overall lens function (transparency, lens shape change) is unclear. However, these relationships are being elucidated using new imaging methodology and quantitative assessments of lens structural and cellular features2,4,19,20,21,22. New protocols to image whole lenses that allow for high spatial resolution visualization of the lens capsule, epithelial cells, and peripheral fiber cells have been developed. This includes methodology to quantify capsule thickness, cell size, cell nucleus size and circularity, meridional row order, fiber cell packing, and fiber cell widths. These visualization and image quantification methods allow in-depth morphometric examination and have advantages over other visualization methods (imaging of flat mounts or tissue sections) by preserving overall 3D tissue structure. These methods have permitted for the testing of novel hypotheses and will enable continued advancement in understanding of lens cell pattern development and function.
For the following experiments, we use wild-type and Rosa26-tdTomato mice tandem dimer-Tomato (B6.129(Cg)-Gt(ROSA) (tdTomato)23 (Jackson Laboratories) in the C57BL/6J background between the ages of 6 and 10 weeks, of both sexes. The tdTomato mice allow for visualization of cellular plasma membranes in live lenses via expression of tdTomato protein fused to the N-terminal 8 amino acids of a mutated MARCKS protein that targets the plasma membrane via N-terminal myristylation and internal cysteine-palmitoylation sites23. We also use NMIIAE1841K/E1841K mice24 obtained originally from Dr. Robert Adelstein (National Heart, Lung, and Blood Institute, National Institutes of Health, Bethesda, MD). As described previously20, NMIIAE1841K/E1841K&#160;mice in FvBN/129SvEv/C57Bl6 background that has loss of CP49 beaded intermediate filament protein (maintains mature fiber cell morphology and whole lens biomechanics), are backcrossed with C57BL6/J wild-type mice. We screened the offspring for the presence of the wild-type CP49 allele. 
Confocal imaging was performed on a laser-scanning confocal fluorescence microscope with a 20x (NA = 0.8, working distance = 0.55mm, 1x zoom), a 40x (NA = 1.3 oil objective, working distance = 0.2mm, 1x zoom), or a 63x (NA = 1.4 oil objective, working distance = 0.19mm, 1x zoom) magnification. All images were acquired using a pinhole size, which is a determinant of optical section thickness, to 1 Airy Unit (the resultant optical thicknesses are stated in figure legends). Images were processed on Zen Software. Images were exported to .tif format and then imported into FIJI ImageJ Software (imageJ.net).

Nuestro objetivo es determinar los mecanismos moleculares que establecen la intrincada arquitectura de la lente y cómo esta arquitectura establecida regula la función de la lente en la transparencia y el cambio de forma de la lente. Para avanzar en la investigación en nuestro campo, utilizamos nuevos métodos de imagen que nos permiten visualizar las características de la lente con alta resolución espacial, y esto nos permite realizar un análisis cuantitativo de imágenes de las estructuras de la lente y las características celulares. La obtención de imágenes de montaje completo es ventajosa sobre la visualización de secciones de tejido o procedimientos de montaje plano, ya que permite la preservación de la estructura general del tejido en 3D.Esto nos permite realizar un examen morfométrico en profundidad y una cuantificación de la estructura nativa de la lente. El cristalino es un tejido biológico integrado con funciones especializadas que dependen de la localización y las geometrías dependientes de la profundidad de las células y sus estructuras asociadas. El uso de los protocolos de imagen y los métodos de cuantificación demostrados, permitirá una mayor comprensión de cómo se establecen las estructuras de la lente y la compleja organización de la lente.
