Les cellules stromales dans les organes hématopoïétiques sont essentielles à la production normale de sang et les perturbations du stroma hématopoïétique peuvent entraîner de graves altérations de l’entretien hématopoïétique et de la réponse au stress 9,23,24. La connaissance des cellules stromales hématopoïétiques est essentielle pour comprendre les maladies hématologiques ...

Chez l’adulte en bonne santé, la production de novo de cellules sanguines se produit dans la moelle osseuse et le thymus. Les cellules stromales à ces sites sont essentielles au maintien de l’hématopoïèse, mais le stroma constitue moins de 1% du tissu 1,2,3,4. L’obtention d’isolats purs d’hématopoïèse soutenant le stroma constitue donc un défi important, en particulier pour la multiomique unicellulaire qui nécessite un traitement rapide pour obtenir des échantillons de haute qualité. Les composants de différents cocktails de digestion peuvent interférer avec certaines étapes de l’analyse multiomique 5,6. Les protocoles présentés ici détaillent l’isolement d’une grande variété de cellules stromales à partir de la moelle osseuse et des tissus thymiques.
Les perturbations des constituants stromaux dans la moelle osseuse et le thymus entraînent une perturbation profonde du développement des cellules sanguines et peuvent entraîner des tumeurs malignes 7,8,9. L’hématopoïèse soutenant le stroma est endommagée à la suite d’un conditionnement cytotoxique et d’une greffe de moelle osseuse, ce qui entraîne une réduction de la sécrétion de cytokines et de facteurs de croissance qui soutiennent les cellules souches et progénitrices hématopoïétiques (HSPC)2,10,11. De plus, le vieillissement affecte les cellules stromales de la moelle osseuse et du thymus, contribuant probablement aux phénotypes hématopoïétiques âgés. Le thymus est le premier organe à subir une involution importante associée à l’âge. La graisse et le tissu fibreux commencent à remplacer le stroma de soutien des lymphocytes T dès le début de la puberté12,13. Dans la moelle osseuse, la teneur en adipocytes augmente avec l’âge et les niches vasculaires et endostatives sont significativement remodelées 14,15,16.
Pour permettre l’étude du stroma de soutien à l’hématopoïèse dans plusieurs états de stress et dans le cas du thymus des tissus humains et murins, nous avons optimisé les protocoles de digestion précédemment publiés 1,2,8,17,18. Ces protocoles garantissent une isolation efficace et reproductible des cellules, et ils sont compatibles avec le séquençage de l’ARN unicellulaire (scRNAseq) et d’autres types de multiomique.

Le séquençage unicellulaire a permis de cartographier des populations cellulaires hétérogènes dans le stroma des organes hématopoïétiques. Ces méthodologies fournissent une lentille à travers laquelle étudier l’hétérogénéité jusqu’alors non résolue à l’état d’équilibre, ainsi que les changements dans la représentation du type cellulaire induits par des stress extrinsèques ou au cours du vieillissement. Ici, nous présentons des protocoles par étapes pour l’isolement de populations de cellules stromales de haute qualité à partir du thymus murin et humain, ainsi que de l’os et de la moelle osseuse murins. Les cellules isolées par ces protocoles conviennent à la génération d’ensembles de données multiomiques unicellulaires de haute qualité. Les impacts de la digestion des échantillons, de l’appauvrissement de la lignée hématopoïétique, de l’analyse/tri FACS et de la façon dont ces facteurs influencent la compatibilité avec le séquençage unicellulaire sont discutés ici. Avec des exemples de profils FACS indiquant une dissociation réussie et inefficace et des rendements de cellules stromales en aval dans l’analyse post-séquençage, des indications reconnaissables pour les utilisateurs sont fournies. La prise en compte des exigences spécifiques des cellules stromales est cruciale pour obtenir des résultats de haute qualité et reproductibles qui peuvent faire progresser les connaissances dans le domaine.

In the healthy adult, de novo production of blood cells occurs in the bone marrow and the thymus. Stromal cells at these sites are essential for maintenance of hematopoiesis, but stroma constitutes less than 1% of the tissue1,2,3,4. Obtaining pure isolates of hematopoiesis supporting stroma therefore constitutes a significant challenge, particularly for single-cell multiomics that requires expedient processing to obtain samples of high quality. Components of different digestion cocktails may interfere with certain steps in multiomics analysis5,6. The protocols presented here detail the isolation of a wide variety of stromal cells from bone marrow and thymic tissues.
Perturbations of stromal constituents in both bone marrow and thymus result in profound disruption in blood cell development and can result in malignancies7,8,9. Hematopoiesis supporting stroma is damaged following cytotoxic conditioning and bone marrow transplantation, resulting in reduced secretion of cytokines and growth factors that sustain hematopoietic stem and progenitor cells (HSPCs)2,10,11. Furthermore, aging affects bone marrow and thymus stromal cells likely contributing to aged hematopoietic phenotypes. The thymus is the first organ to undergo extensive age-associated involution. Fat and fibrotic tissue start replacing T cell supportive stroma as early as the onset of puberty12,13. In the bone marrow, adipocyte content increases with age and the vascular and endosteal niches are significantly remodeled14,15,16.
To enable study of hematopoiesis supportive stroma across multiple stress states and in the case of the thymus of both human and murine tissue, we have optimized previously published digestion protocols1,2,8,17,18. These protocols ensure efficient and reproducible isolation of cells, and they are compatible with single-cell RNAsequencing (scRNAseq) and other types of multiomics.

Le développement des cellules hématiques dans la moelle osseuse et le thymus dépend fortement du soutien des cellules stromales. Les perturbations de la fonction des cellules stromales hématopoïétiques peuvent entraîner une production sanguine inefficace, voire maligne. Notre laboratoire se concentre sur la compréhension de la régulation de l’hématopoïèse par les cellules stromales et sur la façon dont elle peut être manipulée pour obtenir des avantages thérapeutiques.L’avènement du séquençage de cellules uniques a entraîné une cascade d’avancées récentes dans de nombreux domaines de recherche. C’est également le cas dans notre domaine où la résolution d’une seule cellule a permis d’identifier des populations hétérogènes de cellules stromales dans des organes hématopoïétiques jusqu’alors inconnus. Nos protocoles sont optimisés pour y remédier, et nous espérons qu’ils permettront d’approfondir la recherche sur les cellules stromales afin de s’attaquer à des questions cruciales dans le domaine.
