Vores forskning sigter mod at undersøge hjertesvigt med bevaret ejektionsfraktion, kendt som HFpEF, specifikt undersøge virkningerne af lipotoksicitet. Vi søger at belyse det indviklede forhold mellem metabolisk syndrom og lipotoksicitet i at drive HFpEF-progression og afdække de underliggende mekanismer, der driver HFpEF-patofysiologi ved hjælp af denne eksperimentelle murin model. På trods af den stigende forekomst og begrænsede behandlingsmuligheder for HFpEF, er dets patofysiologiske mekanismer, hovedsageligt relateret til metabolisk syndrom, stadig dårligt forstået.
Denne model giver en unik mulighed for at bygge bro over denne kløft og potentielt udvikle nye terapier til HFpEF. Vores models vigtigste fordel er dens evne til at efterligne betydelig lipidfejlhåndtering, der er observeret hos HFpEF-patienter. Denne musemodel giver værdifuld indsigt i patofysiologien af HFpEF, især hjertemetabolisme.
Vores resultater understreger vigtigheden af fejlhåndtering af fedtsyrer og kolesterol i kardiometabolisk HFpEF. Denne model viser perfekt, hvordan fedtsyremangel forstyrrer stofskiftet på forskellige niveauer. Det giver forskere potentielle muligheder for at opdage nye fælder for HFpEF-patienter.
Her på Shahada Laboratory vil vi fokusere på at oversætte denne model til store dyr. Vi vil undersøge spørgsmål relateret til patogenesen, pludselig død og potentielle terapeutiske interventioner rettet mod lipotoksicitet, inflammation og hjerteremodellering.
