Vår forskning tar sikte på å undersøke hjertesvikt med bevart ejeksjonsfraksjon, kjent som HFpEF, spesielt undersøke effekten av lipotoksisitet. Vi søker å belyse det intrikate forholdet mellom metabolsk syndrom og lipotoksisitet for å drive HFpEF-progresjon og avdekke de underliggende mekanismene som driver HFpEFs patofysiologi ved hjelp av denne eksperimentelle murinmodellen. Til tross for den økende forekomsten og begrensede behandlingsalternativer for HFpEF, er dets patofysiologiske mekanismer hovedsakelig relatert til metabolsk syndrom fortsatt dårlig forstått.
Denne modellen gir en unik mulighet til å bygge bro over dette gapet og potensielt utvikle nye terapier for HFpEF. Modellens viktigste fordel er dens evne til å etterligne betydelig feilhåndtering av lipider observert hos HFpEF-pasienter. Denne musemodellen gir verdifull innsikt i patofysiologien til HFpEF, spesielt hjertemetabolisme.
Våre funn understreker viktigheten av feilhåndtering av fettsyrer og kolesterol i kardiometabolsk HFpEF. Denne modellen viser perfekt hvordan fettsyremangel forstyrrer stoffskiftet på forskjellige nivåer. Det gir forskere potensielle veier for å oppdage nye feller for HFpEF-pasienter.
Her på Shahada-laboratoriet vil vi fokusere på å oversette denne modellen til store dyr. Vi vil undersøke spørsmål knyttet til patogenesen, plutselig død og potensielle terapeutiske intervensjoner rettet mot lipotoksisitet, betennelse og hjerteremodellering.
