Vår forskning syftar till att undersöka hjärtsvikt med bevarad ejektionsfraktion, så kallad HFpEF, och specifikt undersöka effekterna av lipotoxicitet. Vi strävar efter att belysa det intrikata förhållandet mellan metabolt syndrom och lipotoxicitet för att driva HFpEF-progression och avslöja de underliggande mekanismerna som driver HFpEF-patofysiologi med hjälp av denna experimentella murina modell. Trots den ökande prevalensen och de begränsade behandlingsalternativen för HFpEF är dess patofysiologiska mekanismer, främst relaterade till metabolt syndrom, fortfarande dåligt kända.
Denna modell ger en unik möjlighet att överbrygga denna klyfta och potentiellt utveckla nya terapier för HFpEF. Den viktigaste fördelen med vår modell är dess förmåga att efterlikna den betydande felhantering av lipider som observerats hos HFpEF-patienter. Denna musmodell ger värdefulla insikter i patofysiologin för HFpEF, särskilt hjärtmetabolism.
Våra resultat understryker vikten av felaktig hantering av fettsyror och kolesterol vid kardiometabol HFpEF. Denna modell visar perfekt hur fettsyrabrist stör ämnesomsättningen på olika nivåer. Det ger forskare potentiella vägar för att upptäcka nya fällor för HFpEF-patienter.
Här på Shahada-laboratoriet kommer vi att fokusera på att översätta denna modell till stora djur. Vi kommer att undersöka frågor relaterade till patogenesen, plötslig död och potentiella terapeutiska interventioner som riktar sig mot lipotoxicitet, inflammation och hjärtremodellering.
