Bakterieinfektioner är ett stort hot mot människors hälsa. Antimikrobiell resistens ökar och det saknas fortfarande vaccin mot flera viktiga patogener. För att identifiera nya vägar för kontrollstrategier behöver vi veta mer om hur patogener svarar på värdens immunsystem och på antibiotika in vivo i de infekterade vävnaderna.
Bakterier som överlever kan orsaka återfall om antimikrobiella medel inte lyckas utrota en infektion. Med hjälp av serietomografi visar vi att tyfoidfeber som orsakar salmonella kan kvarstå under behandlingen i en specifik del av mjälten. Inflammationen är svag i denna region, vilket stör den kritiska synergin mellan antibiotika och inflammation för effektiv eliminering.
Bakterier är mikrometerstora föremål. Det är svårt att lokalisera sådana små föremål i deras vävnadssammanhang. Seriell tvåfotontomografi kan detektera en enda bakterie i centimeterstora organ och bestämma dess replikationshastighet.
Vi kan också detektera immunceller, till exempel neutrofiler, och bestämma deras rumsliga förhållande till bakterierna. Vi studerar aktiviteten hos bakteriostatiska antibiotika. Dessa antibiotika kan bota infektioner, även om de bara kan hämma bakterietillväxt utan att faktiskt döda patogenen.
Vi bestämmer också spridningen av bakteriella mutanter som saknar vissa virulensfaktorer, och effekten av immundefekter på infektion och behandling.
