Las ADSC son células progenitoras mesenquimales multipotentes con la capacidad de autorrenovarse y diferenciarse en varios linajes celulares. Estas células pueden extraerse de la fracción vascular estromal (FSV) del tejido adiposo durante un procedimiento de lipoaspiración1. Las ADSC se han convertido en un tipo de célula madre ideal para su uso en medicina regenerativa porque estas células son abundantes, mínimamente invasivas para la recolección, de fácil acceso ybien caracterizadas. La terapia con células madre ofrece una posible vía para la cicatrización de heridas al estimular la migración celular, la proliferación, la neovascularización y la reducción de la inflamación dentro de las heridas 3,4. Aproximadamente el 80% de la capacidad regenerativa de las ADSC es atribuible a la señalización paracrina a través de su secretoma5. Anteriormente, se sugirió que una inyección local directa de células madre o factores de crecimiento en el tejido dañado podría provocar suficientes mecanismos de reparación in vivo 6,7,8. Sin embargo, este enfoque enfrentó varios desafíos, como la baja supervivencia y la reducción del injerto de células madre dentro de los tejidos dañados como resultado del ambiente inflamatorio 9. Además, una de las razones citadas fue la falta de una matriz extracelular que apoyara la supervivencia y funcionalidad de las células trasplantadas10. Para superar estos desafíos, ahora se está haciendo hincapié en el desarrollo de portadores de biomateriales para apoyar la viabilidad y función de las células madre.
El cultivo celular tridimensional (3D) mejora la interacción célula a célula y célula a matriz in vitro para proporcionar un entorno que se asemeje mejor al entorno in vivo 11. Los hidrogeles se han estudiado ampliamente como una clase de portadores de biomateriales que proporcionan un entorno 3D para el cultivo de células madre. Estas estructuras están hechas de agua y polímeros reticulados12. La encapsulación de ADSC en hidrogel prácticamente no tiene ningún efecto citotóxico sobre las células durante el cultivo, manteniendo la viabilidad de las células6. Las células madre cultivadas en 3D demuestran una mayor retención de su madre y una mayor capacidad de diferenciación13. Del mismo modo, las ADSC sembradas con hidrogel demostraron una mayor viabilidad y un cierre acelerado de la herida en modelos animales14. Además, la encapsulación de hidrogel aumenta significativamente el injerto y la retención de ADSC en heridas15,16. TrueGel3D está hecho de un polímero, ya sea alcohol polivinílico o dextrano, solidificado por un reticulante, ya sea ciclodextrina o polietilenglicol17. El gel es un hidrogel sintético que no contiene ningún producto animal que pueda interferir con los experimentos o desencadenar una reacción inmune durante el trasplante del gel en un paciente, al tiempo que imita eficazmente una matriz extracelular18. El gel es totalmente personalizable modificando la composición y los componentes individuales. Puede albergar diferentes células madre y apoyar la diferenciación de varios tipos de células ajustando la rigidez del gel19. Los sitios de unión se pueden crear mediante la adición de péptidos20. El gel es degradable por la secreción de metaloproteasas, lo que permite la migración celular21. Por último, es transparente y permite realizar técnicas de imagen.
La PBM es una forma mínimamente invasiva y fácil de realizar de terapia con láser de bajo nivel que se utiliza para estimular los cromóforos intracelulares. Diferentes longitudes de onda provocan diferentes efectos en las células22. La luz en el rango del rojo al infrarrojo cercano estimula el aumento de la producción de trifosfato de adenosina (ATP) y especies reactivas de oxígeno (ROS) al mejorar el flujo a través de la cadena de transporte de electrones23. La luz en los rangos azul y verde estimula los canales iónicos activados por la luz, lo que permite la afluencia inespecífica de cationes, como el calcio y el magnesio, en las células, lo que se sabe que mejora la diferenciación24. three&#45;dimensional cell culture of adipose-derived stem cells in a hydrogel with photobiomodulation augmentation

Las células madre derivadas del tejido adiposo (ADSC), que poseen características mesenquimales multipotentes similares a las células madre, se emplean con frecuencia en medicina regenerativa debido a su capacidad para una amplia gama de diferenciación celular y su capacidad para mejorar la migración, la proliferación y mitigar la inflamación. Sin embargo, las ADSC a menudo enfrentan desafíos en la supervivencia y el injerto dentro de las heridas, principalmente debido a condiciones inflamatorias desfavorables. Aquí, nuestro objetivo fue evaluar el impacto sinérgico de la fotobiomodulación (PBM) en la proliferación y citotoxicidad de ADSC dentro de un marco de cultivo celular en 3D. Las ADSC inmortalizadas se sembraron en hidrogeles de 10 μL a una densidad de 2,5 x 103 células y se sometieron a irradiación utilizando diodos de 525 nm y 825 nm a fluencias de 5 J/cm2 y 10 J/cm2. Se evaluaron los cambios morfológicos, la citotoxicidad y la proliferación a las 24 h y 10 días después de la exposición a PBM. Las ADSC exhibieron una morfología redondeada y se dispersaron por todo el gel como células individuales o agregados esferoides. Es importante destacar que tanto el PBM como el marco de cultivo 3D no mostraron efectos citotóxicos en las células, mientras que el PBM mejoró significativamente las tasas de proliferación de ADSC. ADSCs are mesenchymal multipotent progenitor cells with the capacity to self-renew and differentiate into several cell lineages. These cells can be harvested from the stromal vascular fraction (SVF) of adipose tissue during a lipoaspiration procedure1. ADSCs have emerged as an ideal stem cell type to use in regenerative medicine because these cells are abundant, minimally invasive to harvest, easily accessible, and well characterized2. Stem cell therapy offers a possible avenue for wound healing by stimulating cell migration, proliferation, neovascularization, and reducing inflammation within wounds3,4. Roughly 80% of the regenerative capacity of ADSCs is attributable to paracrine signaling via their secretome5. Previously, it was suggested a direct local injection of stem cells or growth factors into damaged tissue could illicit sufficient in vivo repair mechanisms6,7,8. However, this approach faced several challenges, such as poor survival and reduced stem cell engraftment within damaged tissues as a result of the inflammatory environment 9. Furthermore, one of the reasons cited was a lack of an extracellular matrix to support the survival and functionality of the transplanted cells10. To overcome these challenges, emphasis is now being placed on the development of biomaterial carriers to support stem cell viability and function.
Three-dimensional (3D) cell culture enhances cell-to-cell and cell-to-matrix interaction in vitro to provide an environment that better resembles the in vivo environment11. Hydrogels have been extensively studied as a class of biomaterial carriers that provide a 3D environment for stem cell culture. These structures are made of water and crosslinked polymers12. Encapsulation of ADSCs in hydrogel has virtually no cytotoxic effect on the cells during culture while maintaining the viability of the cells6. Stem cells cultured in 3D demonstrate enhanced retention of their stemness and improved differentiation capacity13. Similarly, hydrogel-seeded ADSCs demonstrated increased viability and accelerated wound closure in animal models14. Furthermore, hydrogel encapsulation significantly increases the engraftment and retention of ADSCs in wounds15,16. TrueGel3D is made of a polymer, either polyvinyl alcohol or dextran, solidified by a crosslinker, either cyclodextrin or polyethylene glycol17. The gel is a synthetic hydrogel that does not contain any animal products that may interfere with the experiments or trigger an immune reaction during the transplantation of the gel into a patient while effectively mimicking an extracellular matrix18. The gel is fully customizable by altering the composition and individual components. It can house different stem cells and support the differentiation of several cell types by adjusting the stiffness of the gel19. Attachment sites can be created through the addition of peptides20. The gel is degradable by the secretion of metalloproteases, allowing for cell migration21. Lastly, it is clear and allows for imaging techniques.
PBM is a minimally invasive and easily performed form of low-level laser therapy used to stimulate intracellular chromophores. Different wavelengths elicit different effects on cells22. Light in the red to near-infrared range stimulates increased adenosine triphosphate (ATP) and reactive oxygen species (ROS) production by enhancing flux through the electron transport chain23. Light in the blue and green ranges stimulates light-gated ion channels, allowing for the non-specific influx of cations, such as calcium and magnesium, into cells, which is known to enhance differentiation24. The net effect is the generation of secondary messengers that stimulate the transcription of factors triggering downstream cellular processes such as migration, proliferation, and differentiation25. PBM can be used to pre-condition cells to proliferate or differentiate before transplanting the cells into an adverse environment, e.g., damaged tissue26. Pre- and post-transplant PBM (630 nm and 810 nm) exposure of ADSCs significantly enhanced the viability and function of these cells in vivo in a diabetic rat model27. Regenerative medicine requires an adequate number of cells for effective repair of tissues28. In 3D cell culture, ADSCs have been associated with slower proliferation rates compared to two-dimensional cell culture6. However, PBM can be used to augment the 3D cell culture process of ADSCs by enhancing viability, proliferation, migration, and differentiation29,30.
