Kryogen elektronmikroskopi er afgørende for at bestemme næratomare strukturer af biologiske makromolekyler. På trods af dens afhængighed af gennemsnittet af adskillige billeder med lavt signal-til-støj, er det optimale antal partikler, der er nødvendige for en bestemt opløsning, stadig ukendt. Denne begrænsning hindrer fremskridt inden for prøveanalyse og forberedelsesmetoder.
For at tackle dette introducerer vi en iterativ sorteringsmetode, CryoSieve. Standardprotokolvalg omfatter todimensionel og tredimensionel klassificering, andre protokolsorteringskriterier såsom den normaliserede krydskorrelationsmetode, vinkelgrafkonsistensmetoden og ikke-justeringsklassificeringen er i øjeblikket i brug. Omfattende eksperimenter viser, at CryoSieve overgår andre cryo-EM-partikelsorteringsalgoritmer, hvilket afslører, at de fleste partikler er unødvendige i endelige stakke.
Mindretallet af partikler, der er tilbage i de endelige stakke, giver meget høj opløsning amplitude i rekonstruktive tæthedskort. For nogle datasæt nærmer størrelsen af den fineste delmængde sig den teoretiske grænse. I kryo-EM hindrer prøveforberedelse arbejdsgangen.
På grund af manglen på standardmålinger til protokolsammenligning kan forholdet mellem udvalgte og indsamlede partikler tjene som en kvalitetsmåling. Undersøgelse af deres rumlige og tidsmæssige fordeling kan også fremhæve vigtige fysiske faktorer i forberedelsens effektivitet.
