Kryogen elektronmikroskopi är avgörande för att bestämma nära-atomära strukturer hos biologiska makromolekyler. Trots att den är beroende av ett medelvärde av många bilder med låg signal-till-brus, är det optimala antalet partiklar som behövs för en specifik upplösning fortfarande okänt. Denna begränsning hindrar framsteg i provanalys- och beredningsmetoder.
För att ta itu med detta introducerar vi en iterativ sorteringsmetod, CryoSieve. Val av standardprotokoll inkluderar tvådimensionell och tredimensionell klassificering, andra protokollsorteringskriterier som den normaliserade korskorrelationsmetoden, vinkeldiagrammets konsistensmetod och non-alignment-klassificeringen används för närvarande. Omfattande experiment visar att CryoSieve överträffar andra algoritmer för sortering av kryo-EM-partiklar, vilket avslöjar att de flesta partiklar är onödiga i slutliga stackar.
Den minoritet av partiklar som finns kvar i de slutliga stackarna ger mycket högupplöst amplitud i rekonstruktiva densitetskartor. För vissa datauppsättningar närmar sig storleken på den finaste delmängden den teoretiska gränsen. I cryo-EM hindrar provberedning arbetsflödet.
På grund av bristen på standardmått för protokolljämförelse kan förhållandet mellan utvalda och insamlade partiklar fungera som ett kvalitetsmått. Att undersöka deras rumsliga och tidsmässiga fördelning kan också belysa viktiga fysiska faktorer för förberedelsens effektivitet.
