Vores forskningsmål er at etablere robuste teknikker til at generere basistarmsmonolagsgrænseflader afledt af voksne kvæg-, små- og stortarmsorganoider. Vi anvendte en multimodal analytisk tilgang til at bekræfte den funktionelle epitelbarrieredannelse inden for de etablerede monolag. I modsætning til tredimensionelle modeller tilbyder 2D-monolag en eksponeret, tilgængelig luminal overflade, der er afgørende for at studere værtspatogeninteraktioner.
Disse monolag, der afspejler in vivo-tarmen med flere cellelinjer, giver nu skræddersyede protokoller til undersøgelse af patogener i specifikke tarmsektioner såsom tynd- og stortarmsnicher. Vores forskning identificerede optimerede dyrkningsbetingelser til at skabe og opretholde 2D-monolag fra celler fra kvæg indkøbscenter og tyktarmsorganoider. Vi bekræftede dannelsen af funktionel epitelbarriere i disse monolag ved hjælp af transepiteliale elektriske registranter og paracellulære permeabilitetsanalyser samt immuncytokemiske farvningsteknikker.
Vores protokol adresserede forskningshullet ved at fokusere på manglen på tarmorganoid afledte monolagsmodeller af kvæg. Disse modeller er blevet godt undersøgt i mus og mennesker, men kvæg, som er vigtige reservoirer for tarmpatogener med alvorlige konsekvenser for folkesundheden, er ikke blevet undersøgt tilstrækkeligt. Vores forskning etablerer og karakteriserer et 2D kvæg monolagskultursystem.
Dette innovative værktøj giver mulighed for at udforske normale og sygdomstilstande i kvægets tarmfysiologi, med potentielle anvendelser, der strækker sig til biomedicinsk og translationel forskning af betydning for folkesundheden.
