Vårt forskningsmål är att etablera robusta tekniker för att generera basinära intestinala monolagergränssnitt som härrör från vuxna nötkreaturs små och stora tarmorganoider. Vi använde ett multimodalt analytiskt tillvägagångssätt för att bekräfta bildningen av den funktionella epitelbarriären inom de etablerade monolagren. Till skillnad från tredimensionella modeller erbjuder 2D-monolager en exponerad, tillgänglig luminal yta som är avgörande för att studera värdpatogeninteraktioner.
Dessa monolager, som återspeglar tarmen in vivo med flera cellinjer, ger nu skräddarsydda protokoll för att undersöka patogener i specifika tarmsektioner som tunn- och tjocktarmsnischer. Vår forskning identifierade optimerade odlingsförhållanden för att skapa och upprätthålla 2D-monolager från celler från bovina köpcentrum och tjocktarmsorganoider. Vi bekräftade bildandet av funktionell epitelbarriär inom dessa monolager med hjälp av transepiteliala elektriska registranter och paracellulära permeabilitetsanalyser, samt immuncytokemiska färgningstekniker.
Vårt protokoll tog itu med forskningsgapet genom att fokusera på bristen på intestinala organoidhärledda monolagermodeller av nötkreatur. Dessa modeller har studerats väl på möss och människor, men nötkreatur, som är stora reservoarer för tarmpatogener med allvarliga konsekvenser för folkhälsan, har inte utforskats tillräckligt. Vår forskning etablerar och karakteriserar ett 2D-odlingssystem med ett enda lager av nötkreatur.
Detta innovativa verktyg gör det möjligt att utforska normala och sjukdomstillstånd i bovint tarmfysiologi, med potentiella tillämpningar som sträcker sig till biomedicinsk och translationell forskning av betydelse för folkhälsan.
