Upptäckten av stamceller från mänskliga embryon (hESCs) har ökat dramatiskt de verktyg som finns för att medicinska forskare intresserade av regenerativ medicin. Molekylär avbildning har gett utredare en hög genomströmning, billig och känsliga metoder för att spåra in vivo celltillväxt under dagar, veckor och även månader. Denna utveckling har avsevärt ökat förståelsen för tid och rum kinetik hESC engraft, proliferation och teratom-formation i levande individer. Ett stort framsteg inom molekylär bildanalys har varit en förlängning av icke-invasiv analyser reporter gen från molekylär-och cellbiologi till in vivo multimodalitet bildbehandling plattformar. Dessa reporter gener, under kontroll av tekniska medel och medel som utnyttjar värdcellens s transkriptionell maskiner införs i celler med hjälp av olika vektor och icke-vektor metoder. Väl inne i cellen, kan reporter gener transkriberas antingen konstitutivt eller endast under särskilda biologiska eller cellulära villkor, beroende på vilken typ av promotor. Transkription och översättning av reporter gener i bioaktiva proteiner sedan detekteras med känslig, icke-invasiv instrumentering (t ex CCD-kameror) med hjälp av signal-genererande sonder som D-luciferin. För att undvika behovet av excitatoriska lätta att spåra stamceller in vivo som krävs för fluorescens avbildning, mareld reporter gen bildsystem behöver endast en exogent administrerad sond för att framkalla ljus utsläpp. Firefly luciferas, som härrör från eldfluga Photinus pyralis, kodar ett enzym som katalyserar D-luciferin till optiskt aktiva metabolit, oxyluciferin.
