Numerous biochemical as well as electrophysiological techniques require tissue that must be retrieved very quickly following death in order to preserve the physiological integrity of the neuronal environment. Therefore, the ability to accurately predict the precise locations of brain regions of interest (ROI) and to retrieve those areas as quickly as possible following the brain harvest is critical for subsequent analyses. One way to achieve this objective is the utilization of high-resolution MRI to guide the subsequent dissections. In the present study, individual MRI images of the brains of rhesus and cynomolgus macaques that had chronically self-administered ethanol were employed in order to determine which blocks of dissected tissue contained specific ROIs. MRI-guided brain dissection of discrete brain regions was completely accurate in 100% of the cases. In comparison, approximately 60-70% accuracy was achieved in dissections that relied on external landmarks alone without the aid of MRI. These results clearly demonstrate that the accuracy of targeting specific brain areas can be improved with high-resolution MR imaging.

Zahlreiche biochemische sowie elektrophysiologische Techniken erfordern Gewebe, das sehr schnell nach Tod abgerufen werden muss, um die physiologische Integrit&auml;t der neuronalen Umwelt zu bewahren. Numerosas t&eacute;cnicas bioqu&iacute;micas como electrofisiol&oacute;gicas requieren tejido que debe ser recuperado muy r&aacute;pidamente tras la muerte con el fin de preservar la integridad fisiol&oacute;gica del ambiente neuronal. Por lo tanto, la capacidad para predecir con precisi&oacute;n la ubicaci&oacute;n precisa de las regiones del cerebro de inter&eacute;s (ROI) y para recuperar esas zonas tan pronto como sea posible despu&eacute;s de la cosecha del cerebro es fundamental para posteriores an&aacute;lisis. Una forma de lograr este objetivo es la utilizaci&oacute;n de MRI de alta resoluci&oacute;n para guiar las disecciones posteriores. En el presente estudio, se emplearon im&aacute;genes individuales de MRI de los cerebros de los macacos rhesus y cangrejeros que cr&oacute;nicamente hab&iacute;an autoadministrable etanol para determinar que bloques de tejido disecado contienen espec&iacute;ficos ROIs. Alcoholism and alcohol use disorders are characterized by several months to decades of heavy and problematic drinking, interspersed with periods of abstinence and relapse to heavy drinking. This alcohol-drinking phenotype was modeled using macaque monkeys to explore neuronal adaptations in the striatum, a brain region controlling habitual behaviors. Prolonged drinking with repeated abstinence narrowed the variability in daily intake, increased the amount of ethanol consumed in bouts, and led to higher blood ethanol concentrations more than twice the legal intoxication limit. After the final abstinence period of this extensive drinking protocol, we found a selective increase in dendritic spine density and enhanced glutamatergic transmission in the putamen, but not in the caudate nucleus. Intrinsic excitability of medium-sized spiny neurons was also enhanced in the putamen of alcohol-drinking monkeys in comparison with non-drinkers, and GABAeric transmission was selectively suppressed in the putamen of heavy drinkers. These morphological and physiological changes indicate a shift in the balance of inhibitory/excitatory transmission that biases the circuit toward an enduring increase in synaptic activation of putamen output as a consequence of prolonged heavy drinking/relapse. The resultant potential for increased putamen activation may underlie an alcohol-drinking phenotype of regulated drinking and sustained intoxication.

Los trastornos de uso de alcohol y el alcoholismo se caracteriza por varios meses a d&#xE9;cadas de consumo excesivo de alcohol y problem&#xE1;ticas, intercalados con per&#xED;odos de abstinencia y reca&#xED;da en el consumo excesivo de alcohol. Este fenotipo de beber alcohol se ha realizado utilizando monos para estudiar las adaptaciones neuronales en el cuerpo estriado, una regi&#xF3;n del cerebro para controlar los comportamientos habituales. Consumo prolongado con la abstinencia repetidos redujo la variabilidad en el consumo diario, el aumento de la cantidad de etanol que se consume en los combates, y dio lugar a una mayor concentraci&#xF3;n de etanol en sangre m&#xE1;s del doble del l&#xED;mite legal de intoxicaci&#xF3;n. Despu&#xE9;s del per&#xED;odo de abstinencia final de este protocolo potable extensa, hemos encontrado un aumento selectivo de la densidad de la espina dendr&#xED;tica y transmisi&#xF3;n glutamat&#xE9;rgica mejorada en el putamen, pero no en el n&#xFA;cleo caudado. La excitabilidad intr&#xED;nseca de las neuronas espinosas de tama&#xF1;o medio se increment&#xF3; tambi&#xE9;n en el putamen de los monos para beber alcohol en comparaci&#xF3;n con los no bebedores, y la transmisi&#xF3;n GABAeric fue reprimida de forma selectiva en el putamen de los grandes bebedores. Estos cambios morfol&#xF3;gicos y fisiol&#xF3;gicos indican un cambio en el equilibrio de la transmisi&#xF3;n inhibitoria / excitatoria que los sesgos del circuito hacia un aumento permanente en la activaci&#xF3;n sin&#xE1;ptica de la producci&#xF3;n putamen como consecuencia del consumo de alcohol excesivo y prolongado / reca&#xED;da. El potencial resultante para la activaci&#xF3;n del putamen mayor puede ser la base un fenotipo de beber alcohol de la bebida regulada y la intoxicaci&#xF3;n constante.
