An assay of gamma-glutamylcysteine synthetase (gamma-GCS) and glutathione synthetase (GS) in crude extracts of cultured cells and tissues is described. It represents a novel combination of known methods, and is based on the formation of glutathione (GSH) from cysteine, glutamate and glycine in the presence of rat kidney GS for the assay of gamma-GCS, or from gamma-glutamylcysteine and glycine for the assay of GS. GSH is then quantified by the Tietze recycling method. Assay mixtures contain the gamma-glutamyl transpeptidase (GGT) inhibitor acivicin in order to prevent the degradation of gamma-glutamylcysteine and of the accumulating GSH, and dithiothreitol in order to prevent the oxidation of cysteine and gamma-glutamylcysteine. gamma-GCS and GS levels determined by this method are comparable to those determined by others. The method is suitable for the rapid determination of gamma-GCS GS in GGT-containing tissues and for the studies of induction of gamma-GCS and GS in tissue cultures.

Representa una novedosa combinaci&oacute;n de m&eacute;todos conocidos y se basa en la formaci&oacute;n de glutati&oacute;n (GSH) de ciste&iacute;na, glutamato y glicina en presencia de ri&ntilde;&oacute;n de rata GS para el ensayo de gamma-GCS, o de gamma-glutamilciste&iacute;na y glicina para el ensayo de g. GSH entonces es cuantificada por el m&eacute;todo de reciclaje de Tietze. Ensayo contiene la gamma-glutamil transpeptidasa (GGT) inhibidor de la acivicin para prevenir la degradaci&oacute;n de gamma-glutamilciste&iacute;na y de la acumulaci&oacute;n de GSH y Ditiotreitol para prevenir la oxidaci&oacute;n de la ciste&iacute;na y la gamma-glutamilciste&iacute;na. gamma-GCS y niveles de GS determinados por este m&eacute;todo son comparables a los determinados por otros. El m&eacute;todo es adecuado para la determinaci&oacute;n r&aacute;pida de gamma-GCS GS en tejidos que contienen GGT y para los estudios de inducci&oacute;n de gamma-GCS y GS en cultivos de tejido.

Glutathione synthesis and growth properties were studied in the gamma-glutamyl transpeptidase(GGT)-negative, non-tumorigenic rat liver oval cell line OC/CDE22, and in its GGT-positive, tumorigenic counterpart line M22. gamma-Glutamylcysteine synthetase (GGCS) activities were comparable. Growth rates of M22 cells exceeded those of OC/CDE22 cells at non-limiting and limiting exogenous cysteine concentrations. A monoclonal antibody (Ab 5F10) that inhibits the transpeptidatic but not the hydrolytic activity of GGT did not affect the growth rates of OC/CDE22, and decreased those of M22 to the OC/CDE22 level. In GSH-depleted M22, but not in OC/CDE22 cells, the rate and extent of GSH repletion with exogenous cysteine and glutamine exceeded those obtained with exogenous cysteine and glutamate. With Ab 5F10, repletion with cysteine/glutamine was similar to that obtained with cysteine/glutamate. Repletion with exogenous GSH occurred only in M22 cells, and was abolished by the GGT inhibitor acivicin. Repletion with gamma-glutamylcysteine (GGC) in OC/CDE22 was resistant to acivicin whereas that in M22 was inhibited by acivicin. Repletion with exogenous GSH or cysteinylglycine (CG) required aminopeptidase activity and was lower than that obtained with cysteine. Unless reduced, CG disulfide did not support GSH repletion. The findings are compatible with the notions that (i) GGT-catalyzed transpeptidation was largely responsible for the growth advantage of M22 cells at limiting cysteine concentration, and for their high GSH content via the formation of GGC from a gamma-glutamyl donor (glutamine) and cyst(e)ine, and (ii) aminopeptidase/dipeptidase activity is rate-limiting in GSH repletion when GSH or CG serve as cysteine sources.

R&eacute;pl&eacute;tion avec GSH exog&egrave;ne ne survenue que chez les cellules M22 et a &eacute;t&eacute; abolie par l&#39;acivicine d&#39;inhibiteur de la GGT. R&eacute;pl&eacute;tion avec gamma-glutamylcyst&eacute;ine (GGC) en OC/CDE22 r&eacute;sistait &agrave; acivicine alors que cette in M22 est inhib&eacute;e par l&#39;acivicine. R&eacute;pl&eacute;tion exog&egrave;ne GSH ou cyst&eacute;inylglycine (CG) a &eacute;t&eacute; inf&eacute;rieure &agrave; celui obtenu avec la cyst&eacute;ine n&eacute;cessaire aminopeptidase activit&eacute; Moins &eacute;lev&eacute;, disulfure de CG n&#39;a pas appuy&eacute; la r&eacute;pl&eacute;tion GSH. Saziet&agrave; con GSH esogeno si &egrave; verificato solo in cellule M22 e venne abolita con la GGT inibitore della acivicin. Saziet&agrave; con gamma-glutamilcisteina (GGC) in OC/CDE22 era resistente a acivicin mentre quello in M22 &egrave; stato inibito da acivicin. Ripetizione con esogeno GSH o cysteinylglycine (CG) necessaria attivit&agrave; amminopeptidasi ed era inferiore a quello ottenuto con cisteina. A meno che ridotto, disolfuro di CG non supportano saziet&agrave; GSH. I reperti sono compatibili con le nozioni che (i) GGT-catalizzata transpeptidation era in gran parte responsabile per il vantaggio della crescita delle cellule M22 a limitare la concentrazione di cisteina, e per la loro alta GSH contenuti tramite la formazione di GGC da un donatore di gamma-glutamil (glutammina) e cisti (e) ine e (ii) amminopeptidasi/dipeptidasi attivit&agrave; sono limitazione della velocit&agrave; nella ripetizione GSH quando GSH o CG servono come fonti di cisteina.

Tasas de crecimiento de las c&eacute;lulas M22 excedieron los de c&eacute;lulas OC/CDE22 no limitantes y limitar las concentraciones de ciste&iacute;na ex&oacute;gena. Un anticuerpo monoclonal (Ab 5F10) que inhibe la transpeptidatic pero no la actividad hidrol&iacute;tica de GGT no afect&oacute; las tasas de crecimiento de OC/CDE22 y los de M22 disminuido el nivel OC/CDE22. En M22 GSH-agotado, pero no en c&eacute;lulas OC/CDE22, el &iacute;ndice y el grado de repleci&oacute;n del GSH ciste&iacute;na ex&oacute;geno y glutamina excedieron los obtenidos con glutamato y ciste&iacute;na ex&oacute;gena. Con 5F10 Ab, repleci&oacute;n con ciste&iacute;na/glutamina fue similar a la obtenida con ciste&iacute;na/glutamato. Repleci&oacute;n con GSH ex&oacute;geno ocurri&oacute; solamente en c&eacute;lulas M22 y fue suprimida por la acivicin de inhibidor de la GGT. Repleci&oacute;n con gamma-glutamilciste&iacute;na (CGG) en OC/CDE22 era resistente a acivicin mientras en M22 fue inhibida por acivicin. Necesaria la actividad aminopeptidasa y la repleci&oacute;n con ex&oacute;genos GSH o cysteinylglycine (CG) fue menor que la obtenida con ciste&iacute;na. A menos que reduce, disulfuro de CG no apoy&oacute; repleci&oacute;n GSH. A sensitive and rapid method for detecting fluorescent dyes at low concentrations in homogenous solution is experimentally demonstrated. Fluorescent-labeled DNA probes are detected by attaching magnetic beads and applying alternating magnetic field gradient. This condenses the fluorescent probes into a small detection volume and eliminates the scattering noise from solution by synchronous detection. For DNA probes concentration of 1 x 10(-13) M the detection signal was 3.3 times higher than the noise, thereby implying detection sensitivity of 3 x 10(-14) M.

A novel fluorescent resonance energy transfer (FRET)-based probe discriminates the target DNA from the control. When detection is made, the FRET-based probe is cleaved using Taq-polymerase activity and fluorescent light is produced. The biotinylated probes are attached to streptavidin-coupled superparamagnetic beads and are maneuvered into oscillatory motion by applying an alternating magnetic field gradient through two electromagnetic poles. The beads are condensed into the detection area and their movement in and out the orthogonal laser beam produces a periodic fluorescent signal that is demodulated using synchronous detection. 1.9pM of the Ibaraki virus NS3 cDNA was detected in homogeneous solution within 18min without separation or washing steps.

This paper reviews the development of a magnetic modulation biosensing (MMB) system for rapid, simple and sensitive detection of biological targets in homogeneous solution at low concentrations. It relies on condensation and modulation of the fluorescent-labeled probes attached to magnetic beads using an alternating magnetic field gradient. Condensation of the beads from the entire volume increases the signal while modulation separates the signal from the background noise of the non-magnetized solution. We first discuss the motivation and challenges in specific DNA sequences detection as well as current approaches to overcome some of these challenges. We then present the MMB system, DNA detection schemes and magnetic beads manipulation in solution. Rapid detection at sub-picomolar concentrations of fluorescent-labeled probes as well as of coding sequences of the non-structural Ibaraki virus protein 3 (NS3) complementary DNA (cDNA) without any washing or separation step is also reviewed. Finally, we show preliminary results of protein detection using a 'sandwich'-based assay.

Questo articolo esamina lo sviluppo di un sistema di biosensori (MMB) modulazione magnetica per la rilevazione rapida, semplice e sensibile di bersagli biologici in soluzione omogenea a basse concentrazioni. Essa si basa sulla condensazione e della modulazione delle sonde fluorescenti-etichetta allegata ai branelli magnetici mediante un gradiente di campo magnetico alternato. Condensazione delle perline dall&#39;intero volume aumenta il segnale mentre modulazione separa il segnale dal rumore di fondo della soluzione non magnetizzato. Discutiamo in primo luogo la motivazione e le sfide nell&#39;individuazione di sequenze specifiche del DNA cos&igrave; come gli attuali approcci per superare alcune di queste sfide. Vi presentiamo quindi il sistema MMB, schemi di rilevamento del DNA e manipolazione branelli magnetici in soluzione. Rilevamento rapido a concentrazioni sub-picomolar di etichetta fluorescente sonde pure a partire da sequenze di codificazione della proteina non-strutturale Ibaraki virus 3 (NS3) il DNA complementare (cDNA) senza alcun lavaggio o passo di separazione viene anche rivisto. El prop&oacute;sito de la presente revisi&oacute;n es asesorar sistem&aacute;ticamente el m&eacute;dico de la comunidad sobre los ex&aacute;menes esenciales que se requieren para mantener la salud de la mujer. Esta revisi&oacute;n no se refiere a la prueba que debe realizarse durante el embarazo debido a la naturaleza &uacute;nica de tales ex&aacute;menes para la detecci&oacute;n.
