Tyrosine O-sulfation is a post-translational modification mediated by one of two Golgi tyrosylprotein sulfotransferases (TPST-1 and -2) expressed in all mammalian cells. Tyrosine sulfation plays an important role in the function of some known TPST substrates by enhancing protein-protein interactions. To explore the role of these enzymes in vivo and gain insight into other potential TPST substrates, TPST-2-deficient mice were generated by targeted disruption of the Tpst2 gene. Tpst2(+/-) mice appear normal and, when interbred, yield litters of normal size with a Mendelian distribution of the targeted mutation. Tpst2(-/-) mice have moderately delayed growth but appear healthy and attain normal body weight by 10 weeks of age. In contrast to Tpst1(-/-) males that have normal fertility, Tpst2(-/-) males are infertile. Tpst2(-/-) sperm are normal in number, morphology, and motility in normal media and appear to capacitate and undergo acrosomal exocytosis normally. However, they are severely defective in their motility in viscous media and in their ability to fertilize zona pellucida-intact eggs. Adhesion of Tpst2(-/-) sperm to the egg plasma membrane is reduced compared with wild type sperm, but sperm-egg fusion is similar or even increased. These data strongly suggest that tyrosine sulfation of unidentified substrate(s) play a crucial role in these processes and document for the first time the critical importance of post-translational tyrosine sulfation in male fertility.

La sulfatation tyrosine joue un r&ocirc;le important dans le fonctionnement de certains substrats TPST connus en am&eacute;liorant les interactions prot&eacute;ine-prot&eacute;ine. Cependant, ils sont s&eacute;v&egrave;rement d&eacute;fectueuses dans leur motilit&eacute; dans les milieux visqueux et dans leur capacit&eacute; &agrave; f&eacute;conder les &oelig;ufs zone pellucide intactes. Adh&eacute;rence de Tpst2(-/-) sperme &agrave; la membrane plasmique de le &oelig;uf est r&eacute;duite compar&eacute; avec le sperme du type sauvage, mais fusion spermatozo&iuml;de-ovule est similaire ou m&ecirc;me augment&eacute;. Tyrosin O-Sulfatierung ist eine post-translationale Modifikation, vermittelt durch eine der beiden Golgi Tyrosylprotein Sulfotransferasen (TPST-1 und-2) ausgedr&uuml;ckt in allen S&auml;ugetierzellen. Tyrosin Sulfatierung spielt eine wichtige Rolle in der Funktion von einigen bekannten TPST-Substraten durch die St&auml;rkung von Protein-Protein Interaktionen. Um die Rolle dieser Enzyme in-vivo zu erkunden und Einblick in andere potenzielle TPST-Substrate, wurden TPST-2-defizienten M&auml;usen durch gezielte St&ouml;rung im Tpst2-gen erzeugt. Tpst2(+/-) M&auml;use erscheinen normal und wenn Wildschweinbest&auml;nde, Ausbeute W&uuml;rfe von normaler Gr&ouml;&szlig;e mit einem Mendelian Verteilung der zielgerichtete Mutation. Tpst2(-/-) M&auml;use haben m&auml;&szlig;ig verz&ouml;gert Wachstum aber gesund erscheinen und normales K&ouml;rpergewicht durch Alter 10 Wochen erreichen. Im Gegensatz zu Tpst1(-/-)-M&auml;nnchen, die normale Fruchtbarkeit haben, sind die Tpst2(-/-) M&auml;nner unfruchtbar. Tpst2(-/-) Spermien sind normal in Anzahl, Morphologie und Motilit&auml;t in normalen Medien und scheinen capacitate und acrosomal Exozytose normalerweise zu unterziehen. Sie sind jedoch stark Defekt in ihre Motilit&auml;t in viskosen Medien und in ihrer F&auml;higkeit, Zona Pellucida-intakte Eier zu befruchten. Haftung des Tpst2(-/-), die Spermien, die Ei-Plasmamembran reduziert werden im Vergleich zu Wildtyp-Sperma, aber Sperma-Ei-Fusion ist &auml;hnlich oder sogar erh&ouml;ht. Diesen Daten zufolge stark Tyrosin Sulfatierung von nicht identifizierten Substrate(s) spielen eine entscheidende Rolle in diesen Prozessen und Dokument zum ersten Mal die entscheidende Bedeutung von POSTTRANSLATIONELLE Tyrosin Sulfatierung in m&auml;nnliche Fruchtbarkeit.

Gezielte St&ouml;rung des Tyrosylprotein Sulfotransferase-2, ein Enzym, die POSTTRANSLATIONELLE Protein-Tyrosin O-Sulfatierung katalysiert, wird M&auml;nnliche Unfruchtbarkeit.

Tirosina O-solfatazione &egrave; una modifica alberino-di traduzione mediata da uno dei due sulfotransferases di tyrosylprotein di Golgi (TPST-1 e -2) espressa in tutte le cellule di mammiferi. Solfatazione tirosina svolge un ruolo importante nella funzione di alcuni noti substrati TPST migliorando le interazioni della proteina-proteina. Per esplorare il ruolo di questi enzimi in vivo e insight in altri substrati TPST potenziali di guadagno, mouse TPST-2-carenti sono stati generati da perturbazioni mirate del gene Tpst2. Tirosina O-sulfataci&oacute;n es una modificaci&oacute;n post-translacional mediada por uno de dos Golgi tyrosylprotein molecular (TPST-1 y -2) expresan en todas las c&eacute;lulas mam&iacute;feras. Prostasin is a glycosylphosphatidylinositol-anchored serine protease, with epithelial sodium channel activation and tumor invasion suppression activities. We identified the bladder as an expression site of prostasin. In the mouse, prostasin mRNA expression was detected by reverse transcription and real-time polymerase chain reaction in the bladder, and the prostasin protein was localized by immunohistochemistry in the urothelial cells. In mice injected intraperitoneally with bacterial lipopolysaccharide (LPS), bladder prostasin mRNA expression was downregulated, whereas the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), interferon-gamma (IFN-gamma), TNF-alpha, IL-1beta, and IL-6 was upregulated. Viral promoter-driven expression of the human prostasin homolog in the bladder of transgenic mice attenuated the LPS induction of iNOS but did not abolish the induction. LPS induction of COX-2, TNF-alpha, IL-1beta, and IL-6 expression, however, was not reduced by prostasin transgene expression. Liposome-mediated delivery of prostasin-expressing plasmid into mouse bladder produced similar attenuation effects on LPS-induced iNOS expression, while not affecting COX-2 or cytokine induction. Mice receiving plasmid expressing a catalytic mutant prostasin did not manifest the iNOS induction attenuation phenotype. We propose a proteolytic mechanism for prostasin to intercept cytokine signaling during LPS-induced bladder inflammation.

Prostasin es una proteasa de serina glicosilfosfatidilinositol anclada, con sodio epitelial canal activaci&oacute;n y tumor invasi&oacute;n supresi&oacute;n actividades. Identificamos la vejiga como un sitio de expresi&oacute;n de prostasin. En el rat&oacute;n, la expresi&oacute;n del mRNA prostasin fue detectada por transcripci&oacute;n reversa y reacci&oacute;n en cadena de polimerasa en tiempo real en la vejiga, y la prote&iacute;na de prostasin fue localizada por inmunohistoqu&iacute;mica en las c&eacute;lulas uroteliales. En ratones inyectados intraperitonealmente con lipopolisac&aacute;rido bacteriano (LPS), expresi&oacute;n del mRNA de vejiga prostasin fue regulada, mientras que la expresi&oacute;n de la &oacute;xido n&iacute;trico sintasa inducible (iNOS), cyclooxygenase-2 (COX-2), interfer&oacute;n-gamma (IFN-gamma), TNF-alfa, IL-1beta e IL-6 fue el alza. Expresi&oacute;n basadas en promotor viral del hom&oacute;logo humano prostasin en la vejiga de ratones transg&eacute;nicos aten&uacute;a la inducci&oacute;n LPS de iNOS pero no suprimi&oacute; la inducci&oacute;n. Inducci&oacute;n de LPS de la COX-2, TNF-alfa, IL-1beta, y la expresi&oacute;n de IL-6, sin embargo, no se redujo por la expresi&oacute;n del transg&eacute;n de prostasin. Entrega liposoma-mediada del pl&aacute;smido prostasin-expresando en vejiga rat&oacute;n hab&iacute;a producido efectos similares de atenuaci&oacute;n en la expresi&oacute;n de iNOS inducida por LPS, mientras que no afecta a la inducci&oacute;n de la COX-2 o citoquinas. Ratones recibiendo pl&aacute;smido expresando un catal&iacute;tico prostasin mutante no manifest&oacute; el fenotipo de atenuaci&oacute;n de la inducci&oacute;n de iNOS. Proponemos un mecanismo proteol&iacute;tico para prostasin interceptar la citoquina se&ntilde;alizaci&oacute;n durante la inflamaci&oacute;n inducida por LPS de la vejiga.

Prostasin aten&uacute;a la expresi&oacute;n de sintasa inducible del &oacute;xido n&iacute;trico en la inflamaci&oacute;n de la vejiga urinaria inducida por lipopolisac&aacute;rido.

Past work indicated that sperm from mice deficient in the inositol polyphosphate 5-phosphatase Inpp5b have reduced ability to fertilize eggs in vitro and reduced epididymal proteolytic processing of the sperm protein A Disintegrin and A Metalloprotease 2 (ADAM2). On the basis of these data, our central working hypothesis was that reduced ADAM cleavage would correlate with reduced sperm-egg binding and fusion and in turn with reduced male fertility in Inpp5b(-/-) mice. Multiple endpoints of reproductive functions [mating trials, in vitro fertilization (IVF) assays and ADAM2 and ADAM3 cleavage] were investigated on a male-by-male basis, with pair-wise correlation analysis used to assess the relationships between these various parameters. Motile sperm from Inpp5b(-/-) mice showed significantly reduced fertilization of zona pellucida-free eggs due to reduced binding to the egg plasma membrane and subsequent fusion. Localization of a mouse sperm protein required for gamete fusion, IZUMO1, appears normal in Inpp5b-null sperm. To our surprise and differing from previous reports, we found that ADAM cleavage was only modestly impaired in numerous Inpp5b-null males and varied between individual animals. Performance in mating trials also differed from past reports. The pair-wise correlation analysis revealed that ADAM2 and ADAM3 cleavage was positively correlated, suggesting that processing of these proteins occurs by related/identical mechanisms, but otherwise, there were few correlations between the reproductive endpoints examined here. Nevertheless, this work provides detailed analysis of the Inpp5b(-/-) phenotype and also a blueprint for multivariate analysis to examine relationships between molecular characteristics and in vitro and in vivo physiological functions.

M&aacute;s all&aacute; de trabajo indicada que los espermatozoides de ratones deficientes en el polifosfato de inositol 5-fosfatasa Inpp5b han reducido capacidad de fertilizaci&oacute;n huevos procesamiento proteol&iacute;tico epid&iacute;dimo in vitro y la reducci&oacute;n de la prote&iacute;na de la esperma A conclusi&oacute;n y A Metaloproteinasa 2 (ADAM2). Sobre la base de estos datos, nuestra hip&oacute;tesis central del trabajo fue que reducido escote ADAM correlacionar&iacute;an con fusi&oacute;n y Uni&oacute;n de esperma reducida-huevo y alternadamente con la reducci&oacute;n de la fertilidad masculina en ratones Inpp5b(-/-). M&uacute;ltiples criterios de valoraci&oacute;n de las funciones reproductivas [apareamiento ensayos, ensayos de fertilizaci&oacute;n in vitro (FIV) y escote ADAM2 y ADAM3] investigaron sobre una base de hombre por hombre, con an&aacute;lisis de correlaci&oacute;n pares utilizados para evaluar las relaciones entre estos distintos par&aacute;metros. Espermatozoides m&oacute;viles de Inpp5b(-/-) ratones demostraron fertilizaci&oacute;n significativamente reducida de huevos sin zona pel&uacute;cida debido a atar reducido a la membrana plasm&aacute;tica de huevo y posterior fusi&oacute;n. Localizaci&oacute;n de una prote&iacute;na de rat&oacute;n esperma necesaria para la fusi&oacute;n de gametos, IZUMO1, parece normal en esperma de Inpp5b-null. Para nuestra sorpresa y a diferencia de informes anteriores, encontramos que escote ADAM s&oacute;lo modestamente fue deteriorada en numerosos machos Inpp5b-null y vari&oacute; entre los individuos. Rendimiento en pruebas de acoplamiento tambi&eacute;n se diferenci&oacute; de informes anteriores. El an&aacute;lisis de correlaci&oacute;n pares revel&oacute; que escote ADAM2 y ADAM3 se correlacion&oacute; positivamente, sugiriendo que procesamiento de estas prote&iacute;nas se produce por mecanismos relacionados/id&eacute;nticos, pero de lo contrario, hubo algunas correlaciones entre los extremos reproductivas aqu&iacute; examinados. Sin embargo, esta obra ofrece un an&aacute;lisis detallado del fenotipo Inpp5b(-/-) y tambi&eacute;n un plan para an&aacute;lisis multivariado examinar las relaciones entre las caracter&iacute;sticas moleculares y funciones fisiol&oacute;gicas in vitro e in vivo.

An&aacute;lisis multivariado de la funci&oacute;n reproductiva masculina en Inpp5b-/-ratones revelan heterogeneidad en defectos en la fertilidad, interacci&oacute;n de la membrana del espermatozoide-huevo y clivaje proteol&iacute;tico de esperma ADAMs.

Fertilization is a complex process comprised of numerous steps. During fertilization, two highly specialized and differentiated cells (sperm and egg) fuse and subsequently trigger the development of an embryo from a quiescent, arrested oocyte. Molecular interactions between the sperm and egg are necessary for regulating the developmental potential of an oocyte, and precise coordination and regulation of gene expression and protein function are critical for proper embryonic development. The nematode Caenorhabditis elegans has emerged as a valuable model system for identifying genes involved in fertilization and the oocyte-to-embryo transition as well as for understanding the molecular mechanisms that govern these processes. In this review, we will address current knowledge of the molecular underpinnings of gamete interactions during fertilization and the oocyte-to-embryo transition in C. elegans. We will also compare our knowledge of these processes in C. elegans to what is known about similar processes in mammalian, specifically mouse, model systems.

Tyrosine O-sulfation is a post-translational modification catalyzed by two tyrosylprotein sulfotransferases (TPST-1 and TPST-2) in the trans-Golgi network. Tpst2-deficient mice have male infertility, sperm motility defects, and possible abnormalities in sperm-egg membrane interactions. Studies here show that compared with wild-type sperm, fewer Tpst2-null sperm bind to the egg membrane, but more of these bound sperm progress to membrane fusion. Similar outcomes were observed with wild-type sperm treated with the anti-sulfotyrosine antibody PSG2. The increased extent of sperm-egg fusion is not due to a failure of Tpst2-null sperm to trigger establishment of the egg membrane block to polyspermy. Anti-sulfotyrosine staining of sperm showed localization similar to that of IZUMO1, a sperm protein that is essential for gamete fusion, but we detected little to no tyrosine sulfation of IZUMO1 and found that IZUMO1 expression and localization were normal in Tpst2-null sperm. Turning to a discovery-driven approach, we used mass spectrometry to characterize sperm proteins that associated with PSG2. This identified ADAM6, a member of the A disintegrin and A metalloprotease (ADAM) family; members of this protein family are associated with multiple sperm functions. Subsequent studies revealed that Tpst2-null sperm lack ADAM6 and ADAM3. Loss of ADAM3 is strongly associated with male infertility and is observed in knockouts of male germ line-specific endoplasmic reticulum-resident chaperones, raising the possibility that TPST-2 may function in quality control in the secretory pathway. These data suggest that TPST-2-mediated tyrosine O-sulfation participates in regulating the sperm surface proteome or membrane order, ultimately affecting male fertility.

Lack of tyrosylprotein sulfotransferase-2 activity results in altered sperm-egg interactions and loss of ADAM3 and ADAM6 in epididymal sperm.

Tirosina O-sulfataci&oacute;n es una modificaci&oacute;n post-translacional catalizada por dos Sulfotransferasas tyrosylprotein (TPST-1 y TPST-2) en la red de trans-Golgi. Tpst2-deficiente ratones tienen infertilidad, defectos de la motilidad del esperma y posibles anormalidades en las interacciones de membrana de esperma-huevo. Aqu&iacute; los estudios muestran en comparaci&oacute;n con semen de tipo salvaje, menos atar de esperma Tpst2 null a la membrana del huevo, sino m&aacute;s bien estos avances esperma encuadernado a la fusi&oacute;n de la membrana. Resultados similares fueron observados con esperma salvaje-tipo tratada con el anticuerpo anti-sulfotyrosine PSG2. El mayor grado de fusi&oacute;n de esperma-huevo no es debido a una falta de espermatozoides nulo Tpst2 establecimiento de gatillo del bloque de la membrana del huevo de polispermia. Although the general events surrounding fertilization in many species are well described, the molecular underpinnings of fertilization are still poorly understood. Caenorhabditis elegans has emerged as a powerful model system for addressing the molecular and cell biological mechanism of fertilization. A primary advantage is the ability to isolate and propagate mutants that effect gametes and no other cells. This chapter provides conceptual guidelines for the identification, maintenance, and experimental approaches for the study fertility mutants.
