African sleeping sickness is characterized by alterations in rhythmic functions. It is not known if the disease affects the expression of clock genes, which are the molecular basis for rhythm generation. We used a chronic rat model of experimental sleeping sickness, caused by the extracellular parasite Trypanosoma brucei brucei (Tb brucei), to study the effects on clock gene expression. In tissue explants of pituitary glands from Period1-luciferase (Per1-luc) transgenic rats infected with Tb brucei, the period of Per1-luc expression was significantly shorter. In explants containing the suprachiasmatic nuclei (SCN), the Per1-luc rhythms were flat in 21% of the tissues. We also examined the relative expression of Per1, Clock, and Bmal1 mRNA in the SCN, pineal gland, and spleen from control and infected rats using qPCR. Both Clock and Bmal1 mRNA expression was reduced in the pineal gland and spleen following Tb brucei infection. Infected rats were periodic both in core body temperature and in locomotor activity; however, early after infection, we observed a significant decline in the amplitude of the locomotor activity rhythm. In addition, both activity and body temperature rhythms exhibited decreased regularity and "robustness." In conclusion, although experimental trypanosome infection has previously been shown to cause functional disturbances in SCN neurons, only 21% of the SCN explants had disturbed Per1-luc rhythms. However, our data show that the infection overall alters molecular clock function in peripheral clocks including the pituitary gland, pineal gland, and spleen.

La maladie du sommeil africaine est caract&#xE9;ris&#xE9;e par des alt&#xE9;rations de fonctions rythmiques. On ne sait pas si la maladie affecte l&#x26;#39;expression de g&#xE8;nes de l&#x26;#39;horloge, qui sont la base mol&#xE9;culaire pour la g&#xE9;n&#xE9;ration de rythme. Nous avons utilis&#xE9; un mod&#xE8;le de rat chronique de la maladie du sommeil exp&#xE9;rimentale, caus&#xE9;e par le parasite extracellulaire Trypanosoma brucei brucei (Tb brucei), pour &#xE9;tudier les effets sur l&#x26;#39;expression des g&#xE8;nes d&#x26;#39;horloge. Dans des explants de tissus de glandes pituitaires de p&#xE9;riode1-lucif&#xE9;rase (Per1-luc) chez les rats transg&#xE9;niques infect&#xE9;es par la tuberculose brucei, la p&#xE9;riode de Per1-luc expression &#xE9;tait significativement plus courte. Dans des explants contenant le noyau suprachiasmatique (SCN), les rythmes Per1-luc ont &#xE9;t&#xE9; stables dans 21% des tissus. Expression g&#xE8;nes de l&#x26;#39;horloge pendant l&#x26;#39;inflammation chronique induite par l&#x26;#39;infection &#xE0; Trypanosoma brucei brucei chez les rats

Afrikanischen Schlafkrankheit zeichnet sich durch &Auml;nderungen in rhythmische Funktionen. Es ist nicht bekannt, ob die Krankheit die Expression von Genen Uhr, betrifft die molekulare Grundlage f&uuml;r Rhythmus-Generation sind. Wir verwendeten eine chronische Rattenmodell f&uuml;r experimentelle Schlafkrankheit, verursacht durch die extrazellul&auml;re Parasiten Trypanosoma Brucei Brucei (Tb Brucei), um die Auswirkungen auf die Uhr-Genexpression zu studieren. Im Gewebe Explantaten der Hypophyse aus Dauer1-Luciferase (Per1-Luc) transgenen Ratten mit Tb Brucei infiziert war der Zeitraum Per1-Luc Ausdrucks deutlich k&uuml;rzer. Explantaten, enth&auml;lt die gefundenes Kerne (SCN) waren die Rhythmen Per1-Luc Wohnung in 21 % der Gewebe. Wir untersucht auch den relativen Ausdruck Per1, Taktgeber und Bmal1 mRNA in der SCN, Zirbeldr&uuml;se und Milz von Kontrolle und infizierte Ratten mit qPCR. Beide Uhren und Bmal1-mRNA Ausdruck in der Zirbeldr&uuml;se und Milz nach Tb Brucei-Infektion reduziert. Infizierte Ratten wurden regelm&auml;&szlig;ige, sowohl in der K&ouml;rperkerntemperatur lokomotorische Aktivit&auml;t; jedoch fr&uuml;h nach Infektion beobachteten wir einen signifikanten R&uuml;ckgang in der Amplitude der lokomotorischen T&auml;tigkeit-Rhythmus. Dar&uuml;ber hinaus stellte sowohl Aktivit&auml;t als auch K&ouml;rper-Temperatur-Rhythmen, verminderte Regelm&auml;&szlig;igkeit und &quot;Robustheit.&quot; Obwohl experimentelle gei&szlig;eltragenden Infektion zuvor zu Funktionseinbu&szlig;en SCN Neuronen f&uuml;hren gezeigt wurde, hatte nur 21 % der SCN-Explantaten abschlie&szlig;end Per1-Luc Rhythmen gest&ouml;rt. Valproic acid (VPA) is an anticonvulsant used to treat bipolar disorder, a psychiatric disease associated with disturbances in circadian rhythmicity. Little is known about how VPA affects circadian rhythms. The authors cultured tissues containing the master brain pacemaker for circadian rhythmicity, the suprachiasmatic nuclei (SCN), and skin fibroblasts from transgenic PERIOD2::LUCIFERASE (PER2::LUC) mice and studied the effect of VPA on the circadian PER2::LUC rhythm by measuring bioluminescence. VPA (1 mM) significantly phase advanced the PER2::LUC rhythm when applied at a time point corresponding to the lowest (trough, ~ZT 0) PER2::LUC expression but phase delayed the PER2::LUC rhythm when the drug was administered at the time of highest (peak, ~ZT 12) protein expression. In addition, it significantly increased the overall amplitude of PER2::LUC oscillations at time points at or close to ZT 12 but had no effect on period. Real-time PCR analyses on mouse and human fibroblasts revealed that expressions of other clock genes were increased after 2 h treatment with VPA. Because VPA is known to inhibit histone deacetylation, the authors treated cultures with an established histone deacetylation inhibitor, trichostatin A (TSA; 20 ng/mL), to compare the effect of VPA and TSA on molecular rhythmicity. They found that TSA had similar effects on the PER2::LUC rhythm as VPA. Furthermore, VPA and TSA significantly increased acetylation on histone H3 but in comparison little on histone H4. Lithium is another commonly used treatment for bipolar disorder. Therefore, the authors also studied the impact of lithium chloride (LiCl; 10 mM) on the PER2::LUC rhythm. El &#xE1;cido valproico (VPA) es un anticonvulsivo utilizado para tratar el trastorno bipolar, una enfermedad psiqui&#xE1;trica asociada con alteraciones en el ritmo circadiano. Poco se sabe acerca de c&#xF3;mo afecta a los ritmos circadianos de VPA. Los autores tejidos cultivados que contienen el marcapasos cerebral para el maestro de la ritmicidad circadiana, el n&#xFA;cleo supraquiasm&#xE1;tico (SCN), y fibroblastos de la piel de ratones transg&#xE9;nicos per&#xED;odo2 :: luciferasa (PER2 :: LUC) y se estudi&#xF3; el efecto del &#xE1;cido valproico en el ritmo circadiano PER2 :: LUC midiendo bioluminiscencia. VPA (1 mM) significativamente fase avanzada de la PER2 :: ritmo LUC cuando se aplica a un punto en el tiempo correspondiente a la menor (a trav&#xE9;s, ~ ZT 0) :: PER2 expresi&#xF3;n de LUC, pero un retraso de fase del ritmo LUC PER2 :: cuando el f&#xE1;rmaco se administr&#xF3; en el momento de m&#xE1;xima (pico, ~ ZT 12) expresi&#xF3;n de la prote&#xED;na. Adem&#xE1;s, se aument&#xF3; significativamente la amplitud global de PER2 :: oscilaciones LUC en puntos de tiempo en o cerca ZT a 12 pero no tuvo efecto sobre per&#xED;odo. En tiempo real los an&#xE1;lisis de PCR en rat&#xF3;n y fibroblastos humanos revel&#xF3; que las expresiones de los genes del reloj otros se incrementaron despu&#xE9;s de las 2 h de tratamiento con VPA. Debido a VPA es conocida para inhibir la desacetilaci&#xF3;n de histonas, los autores trataron los cultivos con un inhibidor de la desacetilaci&#xF3;n de histonas establecida, tricostatina A (TSA, 20 ng / ml), para comparar el efecto del &#xE1;cido valproico y la TSA en la ritmicidad molecular.
