Wolfberry (fruit of Lycium barbarum Linn) has been known for balancing 'Yin' and 'Yang' in the body, nourishing the liver and kidney, improving visual acuity for more than 2,500 years in oriental countries. The active components in wolfberry include L. barbarum polysaccharide (LBP), zeaxanthine, betaine, cerebroside and trace amounts of zinc, iron, and copper. Each of them confers distinct beneficial effects and together they help to explain widespread use of wolfberry in the eastern world. Earlier study reported the neuroprotective effects of LBP on retinal ganglion cell (RGC) in an experimental model of glaucoma and the underlying in vivo cellular mechanisms of LBP neuroprotection deserve further exploration. In this study, we adopted proteomics, functional genomics, to evaluate pharmacological effects of LBP on the neuronal survival pathways. Among the significantly changed proteins induced by LBP feeding on ocular hypertension (OH) retinas, only proteins in crystallin family were focused in this study. The proteomic results were further confirmed using the Western blotting of the retinas and immunohistochemical staining of the retinal sections. We demonstrated that neuroprotective effect of-wolfberry extract-LBP on the survival of RGCs may be mediated via direct up-regulation of neuronal survival signal betaB2-crystallin.

Le goji (Lycium barbarum fruit de Linn) a &#xE9;t&#xE9; connu pour &#xE9;quilibrer &#xAB;Yin&#xBB; et &#xAB;Yang&#xBB; dans le corps, nourrissant le foie et les reins, l&#x26;#39;am&#xE9;lioration de l&#x26;#39;acuit&#xE9; visuelle pour les plus de 2500 ans dans les pays orientaux. Les composants actifs dans le goji comprennent L. barbarum polysaccharide (LBP), la z&#xE9;axanthine, la b&#xE9;ta&#xEF;ne, c&#xE9;r&#xE9;broside et des traces de zinc, le fer et le cuivre. Chacun d&#x26;#39;entre eux conf&#xE8;re distincts des effets b&#xE9;n&#xE9;fiques et ensemble, ils aident &#xE0; expliquer l&#x26;#39;utilisation g&#xE9;n&#xE9;ralis&#xE9;e de goji dans le monde oriental. &#xC9;tude ant&#xE9;rieure a rapport&#xE9; les effets neuroprotecteurs de LBP sur les cellules ganglionnaires de la r&#xE9;tine (RGC) dans un mod&#xE8;le exp&#xE9;rimental du glaucome et de le sous-jacent in vivo des m&#xE9;canismes cellulaires de la neuroprotection LBP m&#xE9;ritent une exploration plus pouss&#xE9;e. Dans cette &#xE9;tude, nous avons adopt&#xE9; la prot&#xE9;omique, la g&#xE9;nomique fonctionnelle, pour &#xE9;valuer les effets pharmacologiques de la lombalgie sur les voies de la survie neuronale. Parmi les prot&#xE9;ines chang&#xE9; de mani&#xE8;re significative induite par l&#x26;#39;alimentation LBP sur l&#x26;#39;hypertension oculaire (OH) r&#xE9;tines, seules prot&#xE9;ines en famille cristalline ont &#xE9;t&#xE9; orient&#xE9;s dans cette &#xE9;tude. Les r&#xE9;sultats prot&#xE9;omiques ont &#xE9;t&#xE9; confirm&#xE9;es en utilisant le Western blot des r&#xE9;tines et la coloration immunohistochimique des sections r&#xE9;tiniennes. Nous avons d&#xE9;montr&#xE9; que l&#x26;#39;effet neuroprotecteur de-goji-LBP extrait sur la survie des CGR peut &#xEA;tre m&#xE9;di&#xE9;e par up-r&#xE9;gulation directe du signal de survie neuronale betaB2-crystalline.

Wolfberry (Lycium Barbarum Linn Frucht) ist bekannt f&uuml;r den Ausgleich von &quot;Yin&quot; und &quot;Yang&quot; im K&ouml;rper, n&auml;hrende, Leber und Niere, Verbesserung der Sehsch&auml;rfe f&uuml;r mehr als 2.500 Jahren in orientalischen L&auml;ndern. Die aktiven Bestandteile in Bocksdornfrucht geh&ouml;ren L. Barbarum Polysaccharide (LBP), Zeaxanthine, Betain, Cerebroside und Spur Mengen an Zink, Eisen und Kupfer. Jeder von ihnen verleiht verschiedene wohltuende Wirkung und zusammen um weit verbreitete Nutzung von Bocksdornfrucht in der &ouml;stlichen Welt zu erkl&auml;ren. Fr&uuml;heren Studie berichtet die neuroprotektive Wirkungen von LBP auf retinalen Ganglienzellen Zelle (RGC) in einem experimentellen Modell des Glaukoms und die zugrunde liegenden in-vivo zellul&auml;ren Mechanismen der LBP Nervenschutz verdienen weitere Exploration. In dieser Studie haben wir angenommen, Proteomik, funktionelle Genomik, pharmakologische Wirkungen von LBP auf die neuronale &uuml;berleben-Wege zu bewerten. Unter deutlich ver&auml;nderten Proteine induziert durch LBP F&uuml;tterung auf okul&auml;re Hypertonie (OH) Netzhaut konzentrierten sich in dieser Studie nur Proteine aus der Familie Krystallin. Die Proteomic-Ergebnisse wurden weiter best&auml;tigt mit der westlichen Beflecken der Netzhaut und Immunohistochemical Beflecken der Netzhaut Abschnitte. Wir bewiesen, dass neuroprotektive Wirkung von Bocksdornfrucht Extrakt-LBP auf das &Uuml;berleben der Routinggruppenconnectors kann &uuml;ber direkte bis-Regelung der neuronalen &uuml;berleben Signal betaB2-Crystallin vermittelt werden.

Bis-Regelung der Crystallins ist die neuroprotektive Wirkung von Bocksdornfrucht auf &Uuml;berleben der retinalen Ganglienzellen im Rattenmodell okul&auml;re Hypertonie beteiligt.

In questo studio, abbiamo adottato la proteomica, genomica funzionale, per valutare gli effetti farmacologici di LBP su percorsi di sopravvivenza neuronale. Tra le proteine notevolmente cambiate indotte da LBP nutrendosi di ipertensione oculare retine (OH), solo le proteine della famiglia crystallin erano concentrate in questo studio. I risultati di proteomic erano ulteriormente confermati mediante Western blotting della retina e la colorazione immunoistochimica delle sezioni della retina. Abbiamo dimostrato che neuroprotective effetto di wolfberry extract-LBP sulla sopravvivenza di RGCs pu&ograve; essere mediato via diretta su-regolazione della sopravvivenza neuronale segnale betaB2-crystallin.

Wolfberry (fruta de Lycium Barbarum Linn) ha sido conocido por el equilibrio &#x26;#39;yin&#x26;#39; y &#x26;#39;Yang&#x26;#39; en el cuerpo, nutrir el h&#xED;gado y el ri&#xF1;&#xF3;n, la mejora de la agudeza visual de m&#xE1;s de 2.500 a&#xF1;os en los pa&#xED;ses orientales. Los componentes activos de goji incluyen L. polisac&#xE1;rido barbarum (LBP), zeaxantina, beta&#xED;na, cerebr&#xF3;sido y trazas de zinc, hierro y cobre. Cada uno de ellos confiere distintos efectos beneficiosos y juntos ayudan a explicar el uso generalizado de la baya de goji en el mundo oriental. Estudio anterior inform&#xF3; los efectos neuroprotectores de dolor lumbar en c&#xE9;lulas ganglionares de la retina (CGR) en un modelo experimental de glaucoma y la subyacente en vivo los mecanismos celulares de la neuroprotecci&#xF3;n LBP merecen una exploraci&#xF3;n m&#xE1;s profunda. En este estudio, hemos adoptado la prote&#xF3;mica, gen&#xF3;mica funcional, para evaluar los efectos farmacol&#xF3;gicos del dolor lumbar en las v&#xED;as de supervivencia neuronal. Entre las prote&#xED;nas cambiado de manera significativa inducida por dolor lumbar se alimentan de la hipertensi&#xF3;n ocular (OH) retinas, s&#xF3;lo las prote&#xED;nas de la familia cristalina se centraron en el estudio. Los resultados prote&#xF3;micos fueron confirmados usando el Western Blot de las retinas y la tinci&#xF3;n inmunohistoqu&#xED;mica de las secciones de la retina. Hemos demostrado que efecto neuroprotector de la baya de goji-extracto de dolor lumbar en la supervivencia de la CGR puede ser mediada a trav&#xE9;s de la regulaci&#xF3;n directa de la se&#xF1;al de la supervivencia neuronal betaB2-cristalina.
